摘 要:目的:观察长期灌胃大黄对成年雌性大鼠卵巢组织中Bcl-2和Bax蛋白表达的影响,分析大黄对卵巢功能的影响及与用药剂量和时间的关系。方法:雌性大鼠90只,随机分为正常对照组、大黄水提物1.0 g/kg给药30d、40d、50d、60d组和2.0 g/kg给药30d、40d、50d、60d组,灌胃给药,采用HE染色法观察卵巢组织病理变化,免疫组化法检测卵巢Bcl-2和Bax蛋白表达情况。结果:大黄水提物1.0 g/kg 60d组和2.0 g/kg 40d、50d、60d组Bcl-2表达显著低于正常对照组,Bax表达显著高于正常对照组;给药50d、60d、1.0 g/kg和2.0 g/kg剂量组相同给药时间比较Bcl-2、Bax表达差异具有统计学意义。相同给药剂量不同给药时间组比较:1.0 g/kg 30d与60d组Bcl-2、Bax表达差异具有统计学意义,2.0 g/kg 30d与50,60d组,40d与50、60d组比较Bcl-2、Bax表达差异具有统计学意义,其他各组Bcl-2、Bax表达差异无统计学意义。结论:长期应用大黄可不同程度地下调卵巢中Bcl-2表达,上调Bax表达,其影响程度与给药剂量及时间呈正相关,推测长期应用大黄可能会通过Bcl-2/Bax途径诱导卵巢颗粒细胞凋亡,进而影响卵巢功能。Objective: To investigate the long-term effect of water extract of Rhubarb (~) on expressions of Bcl-2 and Bax in ovary of female rats and the relationship between the effect and administration dosage and time. Method: Ninety female rats aged eight weeks were randomly di- vided into nine groups : normal control group , and drug groups which were ig water extract of Rhubarb 1.0 g/kg for 30d, 40d, 50d, 60d and 2.0 g/kg for 30d, 40d, 50d, 60d. The morphological changes in ovary were observed by HE . The expression of the proteins Bcl-2 and Bax on the ovary were detected by immunohistochemistry method. Results: The protein expressions of Bcl-2 and Bax of 1.0 g/kg for 30d,40d,50d groups and 2.0 g/kg 30d group had no significant difference compared with the normal control group, respectively (P 〉 0.05 ). Whereas the expression of Bcl-2 and Bax in 1.0 g/kg 60d group and 2.0 g/kg 40d,50d,60d groups showed significant deviations from those of the normal group, respectively ( P 〈 0.05 ). The comParison of the groups when administered different dosage for the same time: the ex- pression of Bcl-2 was lower when adminidtered 2.0 g/kg for 50d ,60d than adminidtered 1.0 g/kg while the expression of Bax showed the op- posite result( P 〈 0.05). The comparison of the groups when administered the same dosage for different time: the exPression of Bcl-2 was higher while that of Bax was lower when administered 1.0 g/kg for 60d than 30d ( P 〈 O. 05 ) ; the expression of Bcl-2 decreased while that of Bax increased when adminidstered 50d , 60d than 30d , 40d ( P 〈 0.05). Conclusion: Rhubarb can down-regulate the expression of Bcl- 2 protein and up-regulate the expression of Bax protein in female rat ovary, which were positively correlated with dosage and time. Bcl-2/Bax system might be involved in Rhubarb to induce ovarian granulose cell apoptosis and results in ovarian dysfunction.
