HPLC测定首乌藤饮片中大黄素含量测定方法研究

摘　　要：目的建立HPLC测定首乌藤饮片中总大黄素含量的方法。方法采用Agilent Extend-C18（4.6×250mm,5μm）色谱柱,以甲醇-0.1%磷酸溶液（86∶14）为流动相,检测波长为254nm;柱温25℃;流速为1m L·min-1。结果大黄素的线性范围分别为50~150μg·m L-1（R2=0.9997）,平均加样回收率分别为101.25%（RSD=1.58%,n=9）。结论该方法简单、准确度高、重复性好可用于测定首乌藤饮片中大黄素的含量。OBJECTIVE To develop the method for the determination of thumb in Polygonum mutiflorum by HPLC. METHODS The Agilent Extend-Cls column （4.6 × 250mm,5 μm） was used at 25 %： with the mobile phase consisted of methanol-0. 1% phosphoric acid. The detection wavelength was 254nm and the flow rate was 1mL ·min -1. RESULTS The linear range of thumb was 50 - 150μg · mL-1 （ R2 = 0. 9997 ）. The average recoveries was 101.25% （ RSD = 1.58% , n = 9）. CONCLUSION The method is simple, accurate and reproducible, it can be used for determination of the content of thumb in Polygonum mutiflorum.
