摘  要:目的探讨功夫菊酯(lambda-cyhalothrin,LCT)是否因其拟雌激素作用而影响小鼠海马神经突触发育。方法取孕18 d的ICR小鼠胚胎海马神经细胞体外培养7 d,用LCT(0.1、1、10μmol/L)、雌激素(10 nmol/L)、LCT(10μmol/L)加雌激素(10 nmol/L)、LCT(10μmol/L)加ICI182 780(ERα抑制剂,1μmol/L)、雌激素(10 nmol/L)加ICI182 780(1μmol/L)处理24 h,CCK-8法检测细胞活性,免疫荧光细胞化学法观察神经元突起发育和突触后致密蛋白95(post-synaptic density protein 95,PSD95)。结果 LCT或雌激素单独处理可增加海马神经元突起的数目(F=18.16,P<0.001)和长度(F=12.23,P<0.001)以及PSD95的表达(F=12.71,P<0.001),雌激素受体抑制剂ICI182,780可以阻断该作用(F=12.71,P<0.001);LCT与雌激素联合作用干扰了雌激素对神经元突起数目(F=18.16,P<0.001)和PSD95表达的促进作用(F=12.71,P<0.001)。结论 LCT可抑制雌激素对神经突触发育的促进作用。Objective To investigate the estrogenic effect of Lambda-cyhalothrin(LCT) on mouse hippocampal neurons in vitro.Methods The primary hippocampal neurons were derived from 18-day embryonic mouse and cultured for7 days in vitro,and then were exposed to LCT(0.1,1,10μmol / L),estrogen(E2,10 nmol / L),LCT(10 μmol / L) +ICI182,780(1 μmol / L),E2(10 nmol / L) + ICI182,780(1 μmol / L) and LCT(10 μmol / L) + E2(10 nmol / L) for24 hours.CCK-8 kit was used to detect cell viability.The development of neurite and the expression of post-synaptic density protein 95(PSD95) were measured using immunofluorescent staining.Results LCT or E2 could enhance the growth of neurite in amount(F = 18.16,P〈0.001) and length(F = 12.23,P〈0.001),and PSD95 expression significantly(F =12.71,P〈0.001),but the above effect could be blocked by ICI182,780,an ERα antagonist(F = 12.71,P〈0.001).In the presence of LCT,the promotion of E2 on neurodevelopment was blocked significantly in neurite amount(F = 18.16,P〈0.001) and PSD95 expression(F = 12.71,P〈0.001).Conclusions LCT can inhibit the promotion of E2 on neurodevelopment.
