摘 要:目的:通过探讨雷帕霉素对人脐带来源间充质干细胞(UC-MSCs)衰老的影响,推动其临床应用。方法将人脐带间充质干细胞(MSCS)分为对照组及雷帕霉素组。β-半乳糖苷酶染色实验检测UC-MSC细胞内衰老相关β-半乳糖苷酶(SABG)的表达量;细胞计数试剂(CCK8)检测UC-MSC细胞的增殖;Real-time PCR检测UC-MSC细胞中衰老、增殖与干性相关 mRNA的表达。结果与无水乙醇对照组相比,雷帕霉素组细胞β-半乳糖苷酶染色阳性率及染色深度均降低;细胞增殖加快;增殖、干性基因升高同时伴有衰老相关基因的下降。结论雷帕霉素能够减缓人脐带MSCs的衰老。Objective To investigate the effect of Rapamycin in the senescence process of umbilical cord-derived mesenchymal stem cells (UC-MSCs).Methods UC-MSCs were divided into Rapamycin group and absolute alcohol control group.The measure of senescence-associated b-galactosidase(SABG) of UC-MSCs were detected by Beta galactosidase staining reagent.The proliferation performance of UC-MSC were examined by Cell Counting Kit-8(CCK8).The expressions of senescence,proliferation and stemness relative mRNA were examined by real-time PCR. Results Compared with the absolute alcohol control group,The Rapamycin group show a lower positive rate and dyeing depth of Beta galactosidase staining. Rapamycin treated UC-MSCs present a higher rate of proliferation.The expressions of proliferation,stemness related mRNA were elevated, with an decrease of senescence relative mRNA.Conclusion Rapamycin slows down the senescence phenotype.
