摘  要:用不同浓度的新疆细虫草(Ophiocordyceps gracilis)氯仿相和正丁醇相浸膏和发酵菌丝体的醇提物作用于人胃癌AGS细胞24 h和48 h后,用MTT法测定对细胞的抑制率,并在显微镜下观测细胞形态变化。结果表明:新疆细虫草及菌丝体提取物对人胃癌细胞的增殖有抑制作用,且具有浓度依赖性,当菌丝体醇提物、新疆细虫草正丁醇相和氯仿相浓度为400μg/mL,作用于胃癌细胞48 h时,IC_(50)值分别为(82.0±0.1)μg/mL、(134.4±0.1)μg/mL和(36.0±0.1)μg/mL。Ophiocordyceps gracilis fruit bodies and insect host collected from Burqin Forest, Altay, Xinjiang Province, were dried at 50 ℃, ground to a powder and subjected to Soxhlet extraction with six volumes of 95% ethanol. Combined volumes from three extractions were concentrated approximately nine-fold under vacuum and then extracted sequentially with equal volumes of chloroform and n-butyl alcohol to yield preparations OGL and OGZ, respectively. Fungal mycelium isolated from an O. gracilis fruit body was inoculated into a liquid medium containing (/L) 200 g potato, 20 g sucrose and 10 g peptone, and incubated at 19 ℃ with shaking (180 r/min) for 30 d. Mycelial biomass was harvested, dried at 50 ℃ , ground to powder and subjected to Soxhlet extraction with six volumes of 95% ethanol to yield preparation OGPDC. Inhibitory effects following 24 h and 48 h exposure to OGL, OGZ and OGPDC on AGS gastric cancer cell proliferation were determined using the MTT assay, and changes in cell morphology was observed microscopically. All three extracts inhibited AGS proliferation in a concentration dependent manner, and recorded IC50 values for OGL, OGZ and OGPDC were (82.0 ± 0.1) μg/mL, (134.4 ±0. 1) gg/mL and (36.0± 0.1) μg/mL, respectively following 48 h exposure to 400 μg/mL concentrations.
