摘  要:目的研究葫芦素B对神经母细胞瘤SH-SY5Y细胞增殖的影响及其作用的分子机制。方法通过MTT法检测葫芦素B对SH-SY5Y细胞增殖的影响,流式细胞仪检测葫芦素B对SH-SY5Y细胞凋亡和生理水平上活性氧含量、线粒体膜电位的影响。Hoechst 33258染色检测药物处理前后细胞形态的变化。Westernblot检测葫芦素B处理SH-SY5Y细胞24 h前后Bcl-2、Bax、p21、p53等蛋白合成的变化。结果实验浓度范围内,葫芦素B可时间与浓度依赖性地抑制SH-SY5Y细胞增殖;浓度依赖性地诱导SH-SY5Y细胞凋亡、活性氧水平升高和线粒体膜去极化;在蛋白水平上,葫芦素B能诱导凋亡抑制蛋白Bcl-2下调,促凋亡蛋白Bax、p21、p53上调。结论葫芦素B在体外能显著抑制SH-SY5Y细胞增殖,这一影响可能通过诱导细胞凋亡、ROS积累、线粒体膜去极化及其相关蛋白表达变化来实现。Objective To study the effect of cucurbitacin B on the proliferation of SH-SY5 Y cells,and to clarify the molecular mechanism. Methods MTT assay w as used to test the effect of cucurbitacin B on proliferation of SH-SY5 Y cells. Apoptosis,ROS production and mitochondrial membrane potential of SH-SY5 Y cells w ere detected by flow cytometry. Morphological changes of cells w ith or w ithout drug treated w ere observed by Hoechst 33258 staining.The protein expression of Bcl-2,Bax,p21 and p53 w as investigated by w estern blot. Results In the range of experimental dose,cucurbitacin B inhibited the proliferation of SH-SY5 Y cells in dose-dependent and time-dependent manners,and also induced cell apoptosis,promoted ROS production,depolarized mitochondrial membrane potential in SHSY5 Y cells. In the protein level,the decreasing protein expression of Bcl-2,increasing protein expression of Bax,p21 and p53 w ere also observed in SH-SY5 Y cells after cucurbitacin B treatment. Conclusion Cucurbitacin B inhibits the proliferation of SH-SY5 Y cells significantly by inducing cell apoptosis,changing the physiological level and regulating the protein expressions of Bcl-2,Bax,p53 and p21.
