摘  要:【目的】明确引起新疆葡萄黄化类症状的病毒及类病毒种类,分析其单病毒和复合感染情况,为葡萄病毒及类病毒的防治提供重要的理论依据和有效的检测技术。【方法】利用已报道的6对特异性引物(GRSPa V-RSP48/49、GFKV-L630/U279、GLRav2-V2d CPf2/V2CPr1、HSVd-f/r、GYSVd1-f1/r1和GYSVd2-f2/r2)分别对新疆不同地区黄化类症状的葡萄病样进行RT-PCR检测,并对各引物的扩增产物进行序列分析。【结果】3种病毒及3种类病毒(GRSPa V、GFk V、GLRa V-2、GYSVd2、GYSVd1和HSVd)在新疆葡萄中均有不同程度的感染,其感染率分别为36.1%、51.4%、48.6%、37.5%、37.5%、94.4%;72个病样中,2种以上病毒或类病毒复合感染率约为75%,6种病毒或类病毒复合感染率为10%以上。【结论】可侵染新疆葡萄引起黄化类症状的病毒和类病毒可达6种,且复合感染较为普遍。【Objective】To detect the kind of virus and viroid that cause yellow class symptoms in grape,to confirm the virus types and analyze the situation of composite infection. The obtained result might provide valuable information for the establishment of effective control measures and detection system. 【Method】Six pairs of specific primers GRSPa V- RSP48 / 49,GFKV- L630 / U279,GLRav2- V2cpf2 / V2cpr1,GYSVd1- f / r,GYSVd2- f / r,HSVd- f / r were utilized to detect the yellowing class symptom in samples among different grapes in Xinjiang. And sequence analyses of the amplification product of each primer were conducted.【Result】Molecular detection and sequence analysis showed that three viruses and viroids( GRSPa V,GFk V,GLRa V- 2,GYSVd2,GYSVd1 and HSVd) had different levels of infection in Xinjiang grapes,the detection rate of GRSPa V,GFk V,GLRa V- 2,GYSVd1,GYSVd2,and HSVd were 36. 1%,51. 4%,48. 6%,37. 5%,37. 5% and 94. 4%; Among the 72 cases,75% were infected by two or more kinds of virus or viroid,and morethan 10% were infected by six viruses and viroids.【Conclusion】Six kinds of viruses could cause the yellowing class symptom in grapes in Xinjiang and they were widely spread and caused serious damage.
