菊花滑刃线虫的培养条件和繁殖特性

摘　　要：采用胡萝卜和甘薯愈伤组织培养的方法,分别在4个不同培养温度和6个不同培养时间条件下,观察菊花滑刃线虫（Aphelenchoides ritzemabosi（Schwartz,1911）Steiner＆Buhrer,1932）在2种植物愈伤组织上的繁殖特性。结果表明：菊花滑刃线虫在2种植物愈伤组织上培养繁殖的适温为15~25℃,温度达到30℃及以上均不能正常繁殖。在胡萝卜愈伤组织上培养,20℃时第5周繁殖率最大,为3 937.50倍;在甘薯愈伤组织上培养,20℃时第6周繁殖率最大为576.68倍;在相同培养温度和时间条件下,菊花滑刃线虫在胡萝卜愈伤组织上的繁殖率均大于在甘薯愈伤组织上的繁殖率。The reproductive characteristics of Aphelenchoides ritzemabosi（Schwartz）Steiner Buhrer on carrot（Daucus carota）callus and sweet potato（Ipomoea batatas）callus were studied at 4different temperatures and 6different culture times.The results showed that the optimum temperatures of A.ritzemabosi on two calluses in culture were from 15℃ to 25℃ and it could not reproduce at temperatures greater than or equal to 30℃.The maximum reproduction rates of A.ritzemabosi on the carrot callus and on the sweet potato callus were 3 937.50 at 20℃for 5weeks,576.68 at 20℃for 6weeks,respectively.The reproduction rate of A.ritzemabosi on the carrot callus was higher than that on the sweet potato callus at same temperature and with the same culture time.This research provided the scientific basis for the confirmation of the reproductive cycle,life history and occurrence regularity of A.ritzemabosi,and it also provided a practical method for the systematical study of A.ritzemabosi.
