活血潜阳颗粒对SHR大鼠肾局部组织糜酶系统的作用研究

摘  要:目的:通过活血潜阳颗粒对SHR大鼠肾局部组织的糜酶系统作用,探讨活血潜阳法治疗高血压肾病的可能机制,为活血潜阳颗粒的肾靶器官保护的临床运用提供实验依据。方法:42只SPF级19周龄雄性SHR大鼠随机分为活血潜阳颗粒大剂量组、活血潜阳颗粒中剂量组、活血潜阳颗粒小剂量组,糜酶抑制剂组,模型组,8只SPF级19周龄wky大鼠作为正常对照组。活血潜阳颗粒大剂量组、活血潜阳颗粒中剂量组、活血潜阳颗粒小剂量组分别予活血潜阳颗粒清膏灌胃;糜酶抑制剂组给予SBTI 10 mg·kg-1·d-1灌胃,模型组和WKY组双蒸水5 m L·d-1·只-1灌胃,连续4周。检测血压,尿总蛋白,24 h尿蛋白,肾皮质匀浆AngⅡ,RT-PCR检测各肾皮质PAR-2、TGF-β1及chymse mRNA的表达。结果:活血潜阳颗粒可降低血压(P<0.05),以减少SHR尿总蛋白及24 h尿蛋白定量的排出(P<0.05),抑制肾皮质PAR-2、TGF-β1及chymse的mRNA的表达(P<0.05),减少肾局部组织AngⅡ的生成(P<0.05)。结论:活血潜阳颗粒可能是通过减少尿总蛋白和降压以抑制肾局部组织肥大细胞浸润,减少肾局部组织chymase mRNA表达,从而减少PAR-2介导的大分子尿蛋白的排出,延缓肾纤维化。Objective: With the experimental study of the effects of Blood-Activating and Yang-Suppressing Granules on the local tissues chymase system of SHR rats' kidneys to explore the possible mechanism of blood-activating and yang-suppressing method for treating hypertensive nephropathy and provide experimental basis for the clinical application of Blood-Activating and YangSuppressing Granules for the protection of kidney target organs. Methods: 42 male SPF rats of 19 weeks old were randomized into high dosage group of Blood-Activating and Yang-Suppressing Granules,medium dosage group of Blood-Activating and Yang-Suppressing Granules and low dosage group of Blood-Activating and Yang-Suppressing Granules,chymase inhibitor group and model group,and 8 SPF wky rats of 19 weeks old were used as normal control group. The above first three groups were given intragastric administration of the clear paste of Blood-Activating and Yang-Suppressing Granules; chymase inhibitor group were given intragastric administration of SBTI10 mg / kg / d; model group and WKY group were given intragastric administration of double distilled water,for successive four weeks. After that,the blood pressure,urine total protein,24 urine protein,and renal cortex homogenate AngⅡ were tested. And the various renal cortex of PAR-2,TGF-β1 and the expression of chymse mRNA RT-PCR were also detected.Results: Blood-Activating and Yang-Suppressing Granules can lower the blood pressure( P 〈0. 05) To reduce the SHR urine total protein and the quantitative excretion of urine protein of 24 hours( P 〈0. 05),reduce the produce of local tissues AngⅡ( P 〈0. 05). Conclusion: Blood-Activating and Yang-Suppressing Granules may possibly reduce the urine total protein and lower the blood pressure,inhibit the mast cells infiltration of renal local tissues,reduce the expression of chymse mRNA RT-PCR,and thus decrease the excretion of PAR-2 mediated macromolecular urinary protein and prolong the renal fibrosis.
