摘  要:目的:探讨黄精多糖的提取及分离纯化条件,对得到的单一多糖进行分子量测定、单糖组成分析,并对其一级结构进行初步分析。方法:制黄精经石油醚脱脂后,采用水提醇沉,DEAE-cellulose及葡聚糖凝胶(Sephadex G-200)色谱柱分离纯化;高效液相色谱法进行纯度鉴定及分子量测定;气质联用测单糖组成;红外光谱和核磁共振对其结构进行分析。结果:获得一种多糖(PSP-1-A),数均分子量为:1410,重均分子量(Mw)为:8640,分布宽度(Mw/Mn)为:6.12。单糖主要由半乳糖组成,另外含有少量葡萄糖(Glc)、鼠李糖(Rha)、木糖(Xyl)、阿拉伯糖(Ara)及甘露糖(Man),其摩尔比为93.8∶1.4∶1.3∶1.5∶1.1∶0.9。红外及核磁结构分析证明PSP-1-A是由β-1,4-半乳糖(gal)和β-T-半乳糖(gal)构成的线性多糖。结论:PSP-1-A是一种全新单糖组成的相对分子量均一的黄精多糖。Objective: To discuss the extraction,separation and purification conditions of polygonatum sibiricmu polysaccharides,determine the molecular weight of the single polysaccharide,analyze the composition of the monosaccharide,and further preliminarily analyze its primary structure. Methods: Degrease the propared rhizoma polygonati by petroleum ether,apply water solution and alcohol sedimentation,DEAE-cellulose and Sephadex G-200 chromatographic column separation and purification,identify the purity with high performance liquid chromatography( HPLC),determine the molecular weight,detect the composition of monosaccharide with GC-MS,analyze the structure with infrared spectroscopy and nuclear magnetic resonance. Results: One polysaccharide was got with the molecular weight of 1410,the Mw was 8640,and the Mw / Mn was 6. 12. The monosaccharide was mainly composed by galactose and small amount of glc,rha,xyl,Ara and Man,with the molar ratio of 93. 8∶1. 4∶1. 3∶1. 5 ∶1. 1 ∶0. 9.The analysis of infrared spectroscopy and nuclear magnetic resonance indicates that PSP-1-A was the linear polysaccharide composed by β-1,4-gal and β-T-gal. Conclusion: PSP-1-A is a kind of polygonatum sibiricmu polysaccharide composed by new monosaccharide and of uniform relative molecular weight.
