何首乌饮对衰老大鼠精子DNA完整性的影响

摘　　要：目的:通过观察大鼠精子DNA完整性的变化,探讨何首乌饮对衰老大鼠精子质量的影响。方法:雄性Wistar大鼠随机分为青年对照组、自然衰老组、何首乌丸组、何首乌饮1组和2组。采用精子DNA吖啶橙荧光检测试剂盒检测精子DNA完整性,荧光显微镜下观察,头部发绿色荧光的为DNA正常精子,发红色或黄色荧光为DNA异常精子。结果:自然衰老组大鼠头部发绿色荧光精子数量明显减少,发红色或黄色荧光的DNA异常精子显著增多,与青年对照组比较差异具有统计学意义(P<0.01);何首乌饮和何首乌丸干预组头部发绿色荧光的精子比自然衰老组大鼠显著增多(P<0.01,P<0.05),且用药60d何首乌饮组(1组)头部发绿色荧光的精子明显多于其它两个用药组(P<0.01)。结论:何首乌饮能明显改善自然衰老大鼠精子DNA的完整性,提高精子质量。Objective： To investigate the effects of Heshouwuyin on sperm quality of aging rats by observing the changes of DNA integrity of rats＇ sperm. Methods： Male Wistar rats were randomly divided into young control group, natural aging group, Heshouwuwan group, Heshouwuyin group 1 and Heshouwuyin group 2. Sperm DNA acridine orange fluorescent detection kits was used to detect the sperm DNA integrity. Green fluorescence indicated sperms with normal DNA, while red or yellow fluorescence indicated sperms with abnormal DNA. Results： Compared with young control group, the number of sperms with normal DNA of rats in natural aging group significantly reduced（P 〈0.01）. The number of sperms with normal DNA of rats in Heshouwuwan group and 2 Heshouwuyin groups were obviously more than that of rats in natural aging group（P 〈0.01, P0.05）; Moreover, the number of sperms with normal DNA of rats in Heshouwuyin group 1 was obviously more than other 2 drug groups（P 〈0.01）. Conclusions： Heshouwuyin can improve the DNA integrity and quality of sperms of natural aging rats significantly.
