摘 要:目的:研究金匮肾气丸对UUO致肾间质纤维化大鼠模型的影响,探讨金匮肾气丸抗肾间质纤维化作用机制。方法:采用UUO大鼠模型,将雄性SD大鼠75只按照3:1:1的比例随机分为治疗组(45只)、UUO模型组(15只)、假手术组(15只)。治疗组用金匮肾气丸配成溶液,按照高剂量、中剂量、低剂量分为治疗1组、2组、3组灌胃。灌胃后第7天、第14天、第28天取血液标本,测血肌酐、尿素氮,同时分别处死5只大鼠,取结扎侧肾组织,免疫组织化学技术检测各组大鼠肾脏中单核细胞趋化蛋白-1(monocyte chemoattractant protein-1,MCP-1)、α-平滑肌肌动蛋白(alpha-smooth muscle actin,α-SMA)和细胞增殖核抗原(proliferating cell nuclear antigen,PCNA)表达。结果:UUO造模后肾间质MCP-1、α-SMA、PCNA表达明显增加,使用金匮肾气丸治疗后大鼠肾间质MCP-1、α-SMA、PCNA表达较UUO模型组显著减少(P<0.05)。结论:金匮肾气丸可通过降低MCP-1、α-SMA和MCP-1表达,减轻肾间质纤维化。Objective:To study effect of Jinkui Shenqi Pill on UUO induced renal interstitial fibrosis and expolore the resist interstitial fi-brosis mechanism of Jinkui Shenqi Pill against.Methods:The model of UUO rats,male SD rats were randomly divided into treatment group(n =45),UUO model group(1 5),sham operation group(1 5).The treatment group was given Jingui Shenqi Pill solution,in ac-cordance with the high dose,middle dose and low dose divided into treatment group 1 ,group 2 and group 3,gavage.After intragastric ad-ministration on the 7th day and the 1 4 days,28 days to take blood samples,determination of serum creatinine,blood urea nitrogen,also killed 5 rats and ligation of renal tissue and in the kidney of the rats were detected by immunohistochemical technique in monocyte chemotaxis protein-1 (MCP-1 )and alpha smooth muscle actin(alpha-smooth muscle,actin,and alpha SMA)and proliferating cell nu-clear antigen(proliferating cell nuclear antigen,PCNA expression.Results:After UUO rats renal MCP-1 ,alpha SMA and PCNA expres-sion increased significantly,the renal MCP-1 ,alpha SMA and PCNA expression of rats treated with Jinkui Shenqi Pill were decreased significantly compared with UUO model group(P &lt;0.05).Conclusion:Jinguishenqi pill can reduce MCP-1 ,-SMAalpha and MCP-1 ex-pression and reduce interstitial fibrosis.
