复方苦参碱注射液中苦参碱和氧化苦参碱的总含量测定

摘　　要：目的：建立复方苦参碱注射液中苦参碱和氧化苦参碱总含量的测定方法。方法：采用高效液相色谱法，用氨基键合硅胶填充柱（150×4．6 mm，5μm）；流动相：乙腈-无水乙醇-（体积分数）3％磷酸溶液（801010）；流速：1．0 mL·min －1；检测波长：220 nm；柱温：30℃；进样量10μL。结果：苦参碱在0．17～2．08μg 范围内呈良好的线性关系（r ＝0．9998），氧化苦参碱在0．38～4．59μg 范围内呈良好的线性关系（r ＝0．9997），平均回收率为97．86％（n ＝6），相对标准偏差 RSD 为1．53％。结论：该方法操作简单，准确度高，重复性好，可用于复方苦参碱注射液中苦参碱和氧化苦参碱总含量的测定。Objective：To establish a HPLC method for the determination of matrine and oxymatrine in compound matrine injection.Meth-ods：Using high performance liquid chromatography,with the key amino bonded silica gel packed column（1 50 x 4.6 mm,5 m）;mobile phase：acetonitrile anhydrous ethanol-（volume fraction）3% phosphoric acid solution（801 01 0）;flow rate：1 .0 mL·min -1 ,detec-tion wavelength：220 nm,and the column temperature：30 ℃,1 0 μL of sample volume.Results：Matrine showed a good linear relation-ship in 0.1 7 -2.08 g range（r =0.999 8）and oxymatrine showed a good linear relationship in 0.38 -4.59 g range（r =0.999 7）.The average recovery rate was 97.86%（n =6）,relative standard deviation（RSD）for 1 .53%.Conclusion：This method is simple,accurate and reproducible,and can be used for the determination of matrine and oxymatrine in compound matrine injection..
