烟曲霉孢子对人肺泡上皮细胞早期凋亡的影响

摘  要:目的观察烟曲霉孢子对人肺泡上皮细胞早期凋亡的影响。方法TNF—α或星孢霉素(STS)诱导人肺腺癌A549细胞凋亡。用三种浓度(5×10^5、5×10^6、5×10^7)的烟曲霉孢子与上述细胞共培养,流式细胞仪检测细胞凋亡率的变化。结果TNF-α与未刺激空白对照组比较,刺激后A549细胞的早期凋亡率增高[(14.94±0.82)%比(3.26±0.74)%,x^2=9.18,P〈0.05]STS刺激后A549细胞的早期凋亡率也明显增加[(18.10±1.00)%比(3.26±0.74)%,x^2=12.32,P〈0.053,加入不同浓度的烟曲霉孢子后,A549细胞的早期凋亡率均明显下降(x^2=31.23,P〈0.05),细胞的早期凋亡率与烟曲霉孢子浓度呈负相关(rTNF=-0.92,rSTS=-0.94,均P〈0.05)。结论烟曲霉孢子抑制由TNF-α或STS诱导的人肺腺癌A549细胞的早期凋亡;且烟曲霉孢子浓度越高,抑制作用越明显。Objective To observe the impact of Aspergillus fumigatus conidia on the early apoptosis of alveolar epithelial cells. Methods A549 cells was first stimulated with tumor necrosis factor- α (TNF-α) or staurosporine (STS) to induce cell apoptosis, and then co-cultured with Aspergillus fumigatus conidia of three different concentrations (5× 10^5, 5 × 10^6 , 5 × 10^7 ) simultaneously. The changes of the apoptosis rate of A549 cells were detected by flow cytometry. Results The early apoptosis of A549 cells was increased markedly after stimulated with TNF-α ([14. 94 ± 0. 82]% vs [-3. 26 ± 0.74]%, x^2 9.18, P〈0.05) and STS ([18.10±1.00]% vs [3.26±0.74]%, )x^2 =12.32, P〈0.05). When Aspergillus fumigatus conidia of different concentrations were added, the apoptosis rate was decreased (x^2 =31.23, P〈0.05). The early apoptosis of A549 cells was negatively correlated with the concentration of Aspergillus fumigatus conidia ( rTNF = - 0. 92, rTNF = - 0. 94, both P 〈 0. 05 ). Conclusion Aspergillus fumigatus conidia inhibits the early apoptosis of A549 ceils induced by TNF-α or STS and the inhibit effect increases with the concentration of Aspergillus fumigatus conidia.
