摘  要:目的 :研究不同剂量的地塞米松对妊娠大鼠的胎盘、胎鼠肝脏、肺脏的细胞周期动力学及细胞凋亡的影响。方法 :将 40只定时受孕的Wistar大鼠于妊娠 1 5d时随机分为 4组 ,分别给予安慰剂和不同剂量的地塞米松。妊娠 1 7d时随机选取孕鼠胎盘、胎鼠肝脏和肺脏标本各 5个 ,制成单细胞悬液 ,经流式细胞仪分析各组织细胞的周期动力学及细胞凋亡的变化。结果 :在安慰剂组中 ,以胎鼠肝脏的增殖细胞指数最高。在给药组中 ,虽然各组织的反应略有差异 ,但总体趋势相同 ,即增殖细胞指数下降 ,凋亡细胞的比例上升 ,特别是在大剂量应用地塞米松时 ,作用更明显 ;各组织中的增殖细胞指数与凋亡细胞百分比均呈负相关。结论 :地塞米松可抑制活体细胞的增殖 ,诱导细胞凋亡 。Objective: To study the effects of dexamethsone on the cell cycle kinetics, apoptosis in the placenta, fetal liver and lungs of the pregnant rats. Methods: 40 time bred Wistar rats were randomly assigned to one of the four groups, and given either a placebo or various doses of dexamethasone on the 15th gestational day. On day 17, 5 placentas, 5 fetal livers and 5 fetal lungs in each group were obtained randomly through operation, and single cellular suspensions of those tissues were prepared. The cell cycle kinetics and apoptosis were analyzed by Flow Cytometry. Results: The proportion of the cells in proliferous phase in fetal livers was the highest in the three kinds of tissues in the control group. Although the responses to dexamethasone varied in different tissues, a same tendency was seen, the percentage of the proliferotive cells decreased and the proportion of apoptosis cells increased in all tissues, especially in the large dosage group (group D). There were significant negative correlation between the percentage of proliferous cells and the apoptosis cells. Conclusion: Dexamethasone inhibits the proliferation of the cells in the active tissues and induces the apoptosis of the cells in those tissues.
