摘　　要：目的探讨Runx3过表达对CHB患者Th1、Th2型细胞因子表达水平的影响。方法重组慢病毒载体pGC-FU-Runx3与阴性对照慢病毒载体pGC-FU分别转染29例CHB患者外周血CD4+T细胞,收集培养3 d、5 d和7 d的细胞培养上清液,应用ELISA检测Th1型细胞因子IFN-γ、IL-2和Th2型细胞因子IL-4、IL-10的表达水平。结果与pGC-FU转染组比较,pGC-FU-Runx3转染组Th1型细胞因子IFN-γ的表达水平在3 d(P<0.05)、5 d(P<0.01)和7 d(P<0.01)时均明显升高;IL-2的表达水平在3 d时差异无统计学意义(P>0.05),但在5 d(P<0.05)和7 d(P<0.01)时均明显升高。与pGC-FU转染组比较,pGC-FU-Runx3转染组Th2型细胞因子IL-4的表达水平在3 d时差异无统计学意义(P>0.05),但在5 d(P<0.01)和7 d(P<0.05)时均明显降低;IL-10的表达水平在3 d时差异无统计学意义(P>0.05),但在5 d(P<0.05)和7 d(P<0.05)时均明显降低。与pGC-FU转染组比较,pGC-FU-Runx3转染组IFN-γ/IL-4比值在3 d(P<0.01)、5 d(P<0.01)和7 d(P<0.01)时均明显增大。结论 Runx3过表达可以促进CHB患者Th1型细胞因子的分泌,抑制Th2型细胞因子的分泌,使其Th1/Th2失平衡得到改善。Objective To investigate the effect of Runx3 overexpression on the expression of Th1 ,Th2-type cytokines in patients with chronic hepatitis B （CHB）.Methods Peripheral CD4 ＋T cells derived from 29 CHB patients were transfected with the recombinant lentiviral vector （pGC-FU-Runx3）and the negative control lentiviral vector （pGC-FU）,respectively.Cell culture supernatants were collected on day 3,5 and 7.The expression of Th1-type cytokines （IFN-γ,IL-2 ）and Th2-type cytokines （IL-4,IL-10 ）was measured by enzyme-linked immune sorbent assay （ELISA ）, respectively.Results （1 ）Compared with that in pGC-FU transfected group,the expression of IFN-γ in pGC-FU-Runx3 transfected group significantly increased on day 3 （P 〈0.05）,5 （P 〈0.01）and 7 （P 〈0.01）,respectively.Furthermore, the expression of IL-2 in pGC-FU-Runx3 transfected group significantly increased on day 5 （P 〈0.05）and 7 （P 〈0.01 ）. However,there was no difference in IL-2 expression between pGC-FU transfected group and pGC-FU-Runx3 transfected group on day 3 （P 〉0.05）.（2）Compared to that in pGC-FU transfected group,the expression of IL-4 in pGC-FU-Runx3 transfected group significantly decreased on day 5 （P 〈0.01 ）and 7 （P 〈0.05 ）.Nevertheless,there was no difference in IL-4 expression between pGC-FU transfected group and pGC-FU-Runx3 transfected group on day 3 （P 〉0.05）.Moreover, the expression of IL-10 in pGC-FU-Runx3 transfected group significantly decreased on day 5 （P 〈0.05）and 7 （P 〈0.05）. Nonetheless,there was no difference in IL-10 expression between pGC-FU transfected group and pGC-FU-Runx3 transfected group on day 3 （P 〉0.05）.（3）Compared with that in pGC-FU transfected group,the ratio of IFN-γ/IL-4 in pGC-FU-Runx3 transfected group significantly increased on day 3 （P 〈 0.01 ）,5 （P 〈 0.01 ）and 7 （P 〈 0.01 ）, respectively.Conclusion Runx3 overexpression could promote the secretion of Th1-type cytokines while inhibiting the＆amp;nb
