摘 要:目的利用CO I基因进行我国口岸常见蚊种分类鉴定。方法在云南、广东两地采集常见蚊虫,提取单只蚊虫DNA,采用通用引物扩增其一段CO I基因序列。分析基因同源性及进化情况。结果共测定了14种蚊虫的序列。同种不同地理株之间遗传距离在0~2.5%之间,种间遗传距离为6.6%~18.8%。对40只蚊虫的序列进行基因聚类分析,结果发现,首先聚类的是同种不同地理株之间,然后是库蚊属、伊蚊属、阿蚊属分别聚类,然后是3个属聚类为库蚊亚科,按蚊属则聚类为不同的分支,和形态学分类相一致。结论 COI基因可以对常见蚊种进行有效分类鉴定。Objective Identify common mosquitoes based on the CO I gene. Methods Mosquitoes were collected in Yunnan, Guangdong province, and DNA was extracted from single mosquito. Partial CO I gene was amplified by using a pairs of universal primers, the amplieied products were sequenced and analyzed. Results CO I gene of of40 adult mosquitoes belonging to 14 species collected in Pu'er, Yunnan province, and Shenzhen, Guangzhou province was amplified and sequenced. The intraspecific genetic distance was 0-2.5%, the interspecies genetic distance was6.6%-18.8%. It was found that the different geographical strains belonging to the same species were classified into a cluster, strains of species Culex, Aedes, Armigeres were classified into three clusters, and then clustered into the branch of Culex subfamily. Anopheles clustered in a different branch, which was accordance with morphological classification. Conclusion CO I gene could be used to identify common species of mosquito effectively.
