HPLC法测定乙肝宁颗粒中二苯乙烯苷含量

摘　　要：目的：对乙肝宁颗粒中主要活性成分2,3,5,4＇-四羟基二苯乙烯-2-O-β-D-葡萄糖苷（二苯乙烯苷）含量进行测定。方法：采用HPLC法,色谱条件为：十八烷基硅烷键合硅胶为填充剂;乙腈-水（25∶75）为流动相;检测波长为320nm;流速为1.0ml/min;柱温35℃;进样量10μl。结果：二苯乙烯苷在5～5000μg/ml浓度范围内峰吸光度与浓度呈良好的线性关系（R=1）,平均回收率为98.72%。结论：本方法简便可行、准确可靠,可用于制剂的质量控制。Objective To establish a method for the determination of 2,3,5,4-tetrahydroxyl-diphenylethylene-2-o-glucoside（ TDG） in Yiganning granule by HPLC. Methods The HPLC and ODS column（ 250 mm × 4. 6um,5μm） was applied to determination the content,HCN-H2O（ 25 ∶ 75） as the mobile phase. The flow rate was 1. 0 ml / min,and the deterction wavelength was 320 nm. Result The calibration curve of TDG was linear in the range from 5 to 5000μg / ml（ R = 1）,the average recovery rate was 98. 72%. Conclusion the method is simple,rapid,accurate and reliable. It can be used for the content determination of TDG in Yiganning granule.
