摘 要:VDAC(电压依赖性阴离子通道)又称作线粒体孔蛋白,在调节线粒体的代谢和能量功能以及线粒体介导的凋亡中都发挥重要作用。但水稻中VDAC家族的亚细胞定位及其生物学功能尚未清楚。依据NCBI公布的水稻Os VDAC2的ORF序列设计引物,从水稻品种日本晴叶片c DNA中扩增得到目的片段。构建Os VDAC-GFP融合瞬时表达载体,转化水稻原生质体对其进行亚细胞定位。激光共聚焦观察结果表明:水稻Os VDAC2蛋白主要存在于细胞质,为进一步研究其功能奠定了基础。VDAC (voltage-dependent anion channel), also known as mitochondrial porin, plays an important role in the regulation of metabolic and energetic functions of mitochondria, as well in mitoehondria-mediated apoptosis. However, subcellular localization and biological function of VDAC family in rice are largely unknown. In this study, OsVDAC2 gene was amplified from Nipponbare leaves eDNA based on the published ORF sequence on NCBI. Subcellular localization of OsVDAC2 protein was conducted by construction of transient expression vector pM999-OsVDAC2-GFP and transformation of rice protoplasts. The confocal microscopy results showed that OsVDAC2 protein was located mainly in the cytoplasm, and built a good foundation for further research on the function of OsVDAC2.
