摘 要:目的:探讨人工冬虫夏草对博莱霉素诱导大鼠肺纤维化早期肺泡炎阶段的干预作用。方法:SD大鼠随机分为正常组、模型组(两组均NS灌胃)、强的松组(强的松6 mg·kg-1·d-1灌胃)和虫草菌液组(虫草菌液5 g·kg-1·d-1灌胃),每组各10只。气管内注射博莱霉素建模。肺组织标本进行病理形态学观察及肺泡炎评分;肺泡灌洗液(BLAF)进行细胞总计数及炎症细胞分类计数;BLAF上清检测乳酸脱氢酶(LDH)、髓过氧化物酶、嗜酸粒细胞阳性离子蛋白(ECP)、白介素(IL)-5含量;免疫组化检测肺组织肿瘤坏死因子(TNF)-α表达。结果:与正常组比,模型组肺组织肺泡炎评分升高(P<0.01),BLAF细胞总数、中性粒细胞、淋巴细胞、嗜酸粒细胞比例以及BLAF中的LDH、ECP、IL-5含量明显升高(P<0.05),肺组织TNF-α表达增加(P<0.01);与模型组比,强的松和虫草菌液组肺泡炎评分减少(P<0.01),BALF中细胞总数、淋巴细胞、中性细胞和嗜酸粒细胞比例减少以及LDH、ECP、IL-5含量降低(P<0.05),肺组织TNF-α表达下调(P<0.01)。结论:虫草菌液能减轻博莱霉素诱导的大鼠肺纤维化早期肺泡炎,可能是通过下调IL-5、TNF-α表达,减少嗜酸粒细胞浸润而实现的。Objective:To study the effectiveness of cultured Cordyceps sinensis on early experimental pulmonary fibrosis in rats. Methods:The rats were randomly divided into control and model groups(intratracheally instilled normal saline),prednisone group(intratracheally instilled prednisone 6 mg·kg-1·d-1)and cultured Cordyceps sinensis group(intratracheally instilled 5 g·kg-1·d-1),ten in each group. The rats were intratracheally instilled with bleomycin. The bronchoalveolar lavage fluid(BALF)and lung tissue were collected. The total cells and inflammatory cells of BALF were counted. The contents of lactate dehydrogenase(LDH),myeloperoxidase,eosinophil cationic protein(ECP)and interleukin(IL)-5 in BALF were determined. The lung tissue samples were subjected to TNF-α immunohistochemical test. Results:Compared to controlgroup,the alveolitis score in the model group was increased(P0.01). The total number of cells,the percentage of lymphocyte,neutrophils and eosinophils and the LDH,ECP and IL-5 contents in BALF were increased(P0.05). The TNF-α expression in lung tissue increased(P0.01). Compared to model group,the alveolitis scores in the prednisone and Cordyceps sinensis groups were alleviated(P0.01),the total number of cells,the percentage of lymphocyte and the LDH,ECP,IL-5 contents in BALF decreased(P0.05). The TNF-α expression in lung tissue decreased(P0.01). Conclusion:Cultured Cordyceps sinensisn can alleviate pulmonary alveolitis by reducing IL-5,TNF-α and eosinophils infiltration.
