摘 要:目的:观察辛伐他汀对人非小细胞肺癌H460细胞迁移的影响并探讨其作用机制。方法:CCK-8法检测辛伐他汀对H460细胞活力的影响;用划痕实验和transwell实验观察辛伐他汀对H460细胞迁移的影响;Western bolt检测H460细胞MMP2蛋白的表达。结果:辛伐他汀浓度依赖性地抑制H460细胞迁移,1.25、2.5、5μmol/L的辛伐他汀作用H460细胞24 h,其迁移距离为对照组的79.18%、57.48%、45.34%;与对照组相比,穿过transwell小室膜的细胞数显著减少(P=0.013,P=0.001,P<0.001)。随着辛伐他汀作用浓度增加,H460细胞的MMP2表达逐渐降低。结论:辛伐他汀对人非小细胞肺癌H460细胞的迁移有显著抑制作用,该作用可能与其下调MMP2表达有关。AIM: To evaluate the effect of simvastatin on cell migration of non-small cell lung cancer( NSCLC) cell line H460 as well as the underlying mechanisms. METHODS: CCK-8 assay was used to evaluate the effect of simvastatin on cell viability in H460 cells. Wound healing assay and transwell migration assay were used to determine the effect of simvastatin on cell migration in H460 cells.Western blot was used to detect the effect of simvastatin on expression of MMP2 in H460 cells. RESULTS: Simvastatin inhibited migration of NSCLC H460 cells in a does-dependent manner. After treatment for 24 h with different concentrations of simvastatin( 1. 25,2. 5,5 μmol / L),the wound healing with relative migration distance reduced 79. 18%,57. 48%,45. 34%,respectively; and cell numbers of transwell migration were significantly decreased( P= 0. 013,P = 0. 001,P〈0. 001) compared to control group. With an increase of the concentration of simvastatin exposure,the MMP2 expression in H460 cells gradually decreased. CONCLUSION: These observations indicate that simvastatin inhibits migration of NSCLC H460 cells and its mechanism might be related to the inhibition of MMP2 expression.
