摘 要:[目的]采用碘乙酰胺(iodoacetamide,IA)灌胃刺激联合夹尾应激法建立功能性消化不良(functional dyspepsia,FD)大鼠模型,并对其进行初步评价。[方法]18只10日龄SD大鼠随机分为正常组(9只)和模型组(9只)。正常组大鼠每只给予2%蔗糖溶液0.2ml/d灌胃,持续6d;模型组大鼠每只给予0.1%的碘乙酰胺与2%蔗糖的混合液0.2ml/d灌胃,持续6d。灌胃结束后各组大鼠正常饲养至7周龄,7周龄起以郭氏夹尾刺激法对模型组大鼠刺激1周,正常组不予任何干预,直至大鼠8周龄。大鼠8周龄时检测大鼠胃敏感性(以胃扩张刺激时大鼠颈斜方肌肌电均方根变化率为评价指标)及顺应性(以胃扩张刺激时大鼠胃内球囊容积/压力为评价指标),并于术后第二天检测各组大鼠3h摄食量及胃排空率。[结果]在IA灌胃期间(大鼠10日龄至15日龄),模型组大鼠体重增长率较正常组显著降低(P<0.05)。大鼠7周龄时,各组大鼠体重无显著差异(P>0.05)。7周龄开始予夹尾刺激1周后(大鼠8周龄),模型组大鼠体重较正常组显著降低,结果具有统计学意义(P<0.05)。大鼠8周龄时胃敏感性及顺应性检测结果显示:在胃内球囊压力为40-60mmHg时,模型组大鼠与正常组大鼠相比较,颈斜方肌肌电均方根变化率显著升高(P<0.05),胃顺应性显著降低(P<0.05)。胃排空率结果显示:模型组大鼠胃排空率较正常组大鼠显著降低(P<0.05)。[结论]采用IA灌胃法与夹尾应激法相结合的方法建立FD大鼠模型可在一定程度上模拟FD的临床症状及内脏高敏感、胃顺应性降低、胃排空延缓等病理状态。[Objective]To establish a rat model of functional dyspepsia(FD)resulting from iodoacetamide(IA)-treated and tail-squeezed.[Methods]18ten-day-old male rats were randomly divided into two groups.The model rats(n=9)received 0.2mL 0.1%iodoacetamide(IA)in 2%sucrose daily by oral gavages for 6days;controls(n=9)received 2%sucrose.Rats in both groups were then followed to 7weeks at which point the model rats received tail-squeezed for 1week.Then gastric accommodation and acromiotrapezius(a superficial neck muscle)responses to graded gastric balloon distention(GD;10-60 mm Hg)were tested,and gastric motor function was tested after that.[Results]Model rats exhibited symptoms of FD as compared with the control group.The gastric accommodation was significantly reduced and acromiotrape-zius responses to GD were significantly increased in IA-treated rats.Gastric emptying of model rats was significantly lower than controls.However,no significant gastric pathology was found in both groups.[Conclusion]Our results provide a potential model for further study to functional dyspepsia which simulates the pathogenesis and symptoms of this important clinical problem.
