摘  要:目的 比较紫外分光光度法和高效液相色谱法测定角毛壳菌CH-1发酵液卵孢菌素的差异,建立可靠的定量分析发酵液中卵孢菌素含量的方法。方法 建立标准液梯度,紫外分光光度法在302nm处测量吸光度,高效液相色谱法采用梯度洗脱,检测波长290nm,测定峰面积。运用2种方法分别测量发酵液中卵孢菌素的含量。结果 紫外分光光度法在10~100μg/mL范围内线性关系良好(R2=O.9988),回收率95.13%,测得发酵液中卵孢菌素含量为838.50μg/mL;高效液相色谱法在100~600μg/mL范围内线性关系良好(R2=0.9967),回收率97.62%,测得含量为788.81μg/mL。结论 紫外分光光度法和高效液相色谱法测得发酵液中卵孢菌素含量差异显著,高效液相色谱法测量结果更准确。Objective To investigate the quantitative determination of oosporein in the broth of Chaetomiuncupreum CH-1 by using UV spectrophotometry and HPLC. Methods Different concentration gradients ofoosporein were dissolved in Briton-Robinson buffer solutions and the solutions were detected at 302nm by UVspectrophotometry and at 290nm by HPLC with a binary solvent gradient. Then we measured the content of theoosporein in the broth with the two methods. Results The linear range of oosporein was 10-100μg/mL for UVspectrophtometry (R2=0.9988) and the content of oosporein from the broth was 838.50μg/mL. For HPLC analysis,the linear range was 100-600μg/mL (R2=0.9967) and the content of oosporein in the broth was 788.81μg/mL.Conclusion The content of oosporein in the broth detected by UV spectrophometry and HPLC were significantlydifferent(P〈0.05), it was measured more accurately by HPLC.
