摘 要:目的 研究赛洛多辛对左氧氟沙星在慢性细菌性前列腺炎大鼠前列腺纰织中药动学的影响。方法 构处SD大鼠慢性细菌性前列腺炎模型96只,并随机分成对照组(单用左氧氟沙星)和实验组(赛洛多辛和芹氧氟沙星联合用药),每组各48只,分别十给药后0.125、O.25、0.5、1、2、4、8和12h每个时间点处死6只模型大鼠,采集两组人鼠的前列腺制作前列腺组织匀浆,高放液相色谱(High performance liquid chromatography,HPLC)测定组织中左氧氟沙星浓度,使用3p97软件计算相关药动学参数。结果 对照组和实验组的药.时曲线均符合一室模型。计算出的药动学参数如下:t1/2分别为(4.34±0.25)h和(5.02±0.28)h,tpcak分别为(0.73±O.20)h和(0.86±0.05)h,CL分别为(1.74±0.19)mg/h和(0.64±0.06)mg/h,Cmax分别(3.62±0.3)mg/kg和(8.47±O.51)mg/kg,AUC,1,分别为(21.58±2.00)mg·h/kg和(56.63±4.30)mg·h/kg。结论 赛洛多辛能明娃提粕慢性细荫忭前列腺炎模掣_人鼠lj仃列腺组织中左氧氟沙星的药物浓度。Objective To investigate the influence of silodosin on the pharmacokinetics of levofloxacin inrat's chronic bacterial prostatitis tissue. Methods A total of ninety-six SD rats were used to develop the models ofchronic bacterial prostatitis and then they were divided into two groups randomly control group (only treated withlevofloxacin, n=48) and experimental group (treated with both silodosin and levofloxacin, n=48). Six rats from eachgroup were enthanized to collect their prostate samples to make tissue homogenate at each time point: 0.125, 0.25, 0.5,1, 2, 4, 8 and 12h after levofloxacin administration. High performance liquid chromatography (HPLC) was appliedto detect the concentrations of levofloxacin and 3p97 software program was used to calculate the pharmacokineticparameters. Results The levofloxacin concentration-time curves in rats' prostate of the two groups were both fitthe one-compartment model. The major pharmacokinetic parameters of the control group and experimental group asfollows: tl/2 were (4.34±0.25)h and (5.02±0.28)h, tpeak were (0.73±0.20)h and (0.86±0.05)h, CL were (1.74±0.19)mg/h and (0.64±0.06)mg/h, Cax were (3.62±0.3)mg/kg and (8.47±0.51)mg/kg, AUCH2 were (21.58±2.00)mg.h/kg and(56.63±4.30)mg.h/kg, respectively. Conclusion Silodosin can significantly elevate the concentration of levofloxacinin the prostatic tissues of rats with chronic bacterial prostatitis.
