摘 要:目的:观察刺五加皂苷对D-半乳糖所致衰老大鼠学习记忆功能和海马CA1区LTP的影响,探讨其可能的机制。方法:将27只大鼠随机分为对照组、D-半乳糖组和ASS组。D-半乳糖注射建立衰老大鼠模型,ASS组腹腔注射ASS干预。水迷宫检测大鼠的学习记忆能力。电生理实验比较强直刺激前后海马CA1区PS幅度变化。结果:在水迷宫实验中,ASS能明显改善衰老大鼠的学习记忆功能。电生理实验中,模型组强直刺激后平均PS增幅明显低于对照组,ASS组中衰老大鼠的PS增幅提高。结论:ASS能提高衰老大鼠的学习记忆能力,并改善D-半乳糖对大鼠海马CA1区LTP的抑制。Objective: To observe the effects of ASS on learning and memory capability and LTP in hippocampal CA1 area of the aging rats induced by D-galactose. Methods: 27 SD rats were randomized into 3 groups. The models of aging rats were given injecting D-galactose(120mg·kg^-1·d^-1). The rats in the ASS group were treated with ASS solution(5mg·kg^-1·d^-1, IP); the rats in the control group were treated with the same dose of saline. After 42 days, the learning and memory capability were detected in Morris water maze; electrophysiological method was used to observe the LTP in hippocampal CA1 area. Results: The Morris water maze test showed that ASS significantly improved learning and memory performance in aging rats. In the electrophysiological experiments, PS amplitude significantly decreased in model group versus control, and ASS treatment restored PS amplitude. Conclusion: ASS can improve learning and memory ability and LTP in the hippocampal CA1 of aging rats, and this effect could be attributed to improvement of synaptic dysfunction.
