摘 要:在云南蚕区受害桑树的叶片背部采集到一种新的粉虱类害虫,在实验室观察其成虫和卵的形态特征,并结合线粒体细胞色素C氧化酶亚基Ⅰ基因(mtDNA COⅠ)分子标记技术进行分类鉴定。观察采集粉虱样本的成虫及卵粒四周有粉状物和蜡丝包覆,雄成虫阳茎具有一个双钩结构;提取粉虱样本的基因组DNA,以mtDNA COⅠ基因的通用引物扩增、测序,将拼接序列提交NCBI数据库比对,与数据库中双钩巢粉虱同源序列(GenBank登录号:HM150635.1)的相似度为99%,截取657 bp片段序列比对,二者仅存在1个碱基差异。经过对该粉虱样本的形态识别与mtDNA COⅠ基因分子标记鉴别,初步鉴定其为入侵桑树的双钩巢粉虱(Paraleyrodes pseudonaranjae)。A new whitefly pest was collected from the underside of mulberry leaves of damaged mulberry trees in Yunnan sericulture area. This pest was classified and identified through morphological feature observation of adult and egg in laboratory combined with mitochondrial cytochrome C oxidase subunitⅠgene( mtDNA COⅠ) molecular marker technique. Morphological feature observation showed that the adult and eggs of collected samples were surrounded by powder and wax filaments,and the aedeagus of male adults has a double hook structure. Using genomic DNA extracted from samples of whitefly pest as template,mtDNA COⅠ gene was amplified by PCR with universal primers and sequenced. The spliced sequence was submitted to NCBI database for comparative analysis. The result showed that the spliced sequence has99% sequence identity with the homologous sequence from Paraleyrodes pseudonaranjae( Gen Bank accession No.HM150635. 1). There was only one different base between two sequence fragments of 657 bp. Therefore,these insect samples collected from mulberry trees were preliminarily identified as Paraleyrode pseudonaranjae by morphological recognition and mtDNA COⅠ gene molecular marker identification.
