摘 要:刺蛾科昆虫是柞园栎树林中的重要害虫类群。克隆了栎树林中主要刺蛾科昆虫的线粒体细胞色素C氧化酶亚基Ⅰ基因(mtDNA COⅠ,Gen Bank登录号:KP727647~KP727664),以此作为供试刺蛾科昆虫的DNA条形编码基因分析其碱基组成特点和遗传进化规律,探讨将其应用于栎树林中刺蛾科害虫的分类鉴定和系统进化关系研究的可行性。刺蛾科昆虫各物种间的mtDNA COⅠ基因碱基组成差异不明显,碱基T、C、A、G的平均含量分别为38.0%、16.2%、30.7%和15.1%,AT含量为68.7%,显著高于GC含量(31.3%),表现出明显的AT使用趋向性;碱基明显倾向于使用T(AT偏倚度为-0.106 26),具有较弱的C偏好性(GC偏倚度为-0.035 14),不同物种间该基因碱基T使用偏好程度差异较大。刺蛾科昆虫mtDNA COⅠ基因碱基变异率为26.64%,碱基颠换明显高于转换(R值为0.75)。刺蛾科昆虫各物种mtDNA COⅠ基因的平均进化率为11.5%,进化率最大的发生在锯纹岐刺蛾Austrapoda seres与中国扁刺蛾Thosea sinensis间,进化率最小的发生在黄刺蛾Monema flavescens与梨娜刺蛾Narosoideus flavidorsalis之间。在刺蛾科昆虫mtDNA COⅠ基因的系统进化树中,中国绿刺蛾Parasa sinica和中国扁刺蛾Thosea sinensis聚在一起形成一个单系,其他物种聚在一起形成另外一个单系。以上结果表明,mtDNA COⅠ基因可作为DNA条形编码用于栎树林刺蛾科害虫的分类鉴定和系统进化研究。Insetcs of family Limacodidae are major pests damaging oak yard. Mitochondrial cytochrome C oxidase Ⅰgenes( mtDNA CO Ⅰ) were cloned from family Limacodidae in oak field( Gen Bank accession No. KP727647 to KP727664),and were used as DNA barcodes to analyze base composition characteristics and genetic evolution for exploring the possibility of using mtDNA COⅠ to study identification and phylogenetic taxonomy of family Limacodidae in oak forest. The results indicated that the base composition of mtDNA COⅠ gene had no significant difference between various species. The average base content of T,C,A and G in mtDNA COⅠ genes from family Limacodidae was 38. 0%,16. 2%,30. 7% and 15. 1% respectively. The content of AT( 68. 7%) was significantly higher than that of GC( 31. 3%),showing obvious AT bias. The use of base showed an obvious T bias( AT skew was-0. 106 26) and slight C bias( GC skew was-0. 035 14). There was significant difference in T bias between various species. The base variation rate of mtDNA CO Ⅰ genes from family Limacodidae was 26. 64%,with a obviously higher base trans-version than transition( R value was 0. 75). The average genetic distance of mtDNA COⅠ genes from family Limacodidae was 11. 5%. The genetic distance between Austrapoda seres and Thosea sinensis was the highest,and that between Monema flavescens and Narosoideus flavidorsalis was the lowest. The phylogenetic evolution trees based on mtDNA COⅠ genes displayed that Parasa sinica and Thosea sinensis formed a separate clade,and the other species clustered together and formed another separate clade. These results demonstrated that mtDNA COⅠ genes could be used as DNA barcodes in the study of classification,identification and phylogenetic evolution of insect species from the family Limacodidae.
