摘  要:家蚕属于变温动物,环境温度和湿度对其生长发育有很大影响。选择一对高温高湿耐受性差异显著的家蚕品种932和皓月作为试验材料,采用高通量测序的方法,鉴定2个品种5龄幼虫经高温高湿(35℃±0.5℃,RH 95%±2%)处理后中肠组织中转录水平呈现差异的基因,初步分析这些差异基因参与的相关代谢途径,为探究家蚕品种耐高温高湿的分子机制积累基础信息。在正常环境条件下饲养,2个品种的5龄幼虫中肠组织中有552个表达差异显著的基因,这些差异表达基因共分布于44个GO功能注释小类中,与对高温高湿耐受性较差的皓月相比,对高温高湿耐受性较强的932幼虫,其中肠中与神经突触、抗氧化活性、电子载体活性和受体活性相关的基因高表达。与正常环境条件下饲养相比,在高温高湿环境胁迫下饲养,932与皓月的5龄幼虫中肠中分别有303和276个差异表达基因。通过KEGG Pathway显著性富集分析发现,932的差异表达基因在氧化磷酸化、蛋白质消化与吸收、氨基酸代谢及谷胱甘肽代谢等途径显著富集;皓月的差异表达基因在胰腺分泌、蛋白质消化与吸收、细菌与病毒侵染、m TOR信号等途径显著富集。鉴定的中肠差异表达基因可能与高温高湿胁迫下家蚕体内的生理生化反应及代谢变化等有关。The silkworm( Bombyx mori) is a kind of poikilothermic animal,and environmental temperature has significant effect on its growth and development. There is significant difference in tolerance to high temperature and high humidity between silkworm varieties 932 and Haoyue. To provide basic information for exploring the molecular mechanisms of silkworm resistance to high temperature and high humidity,the 5th instar larvae of silkworm varieties 932 and Haoyue( HY) were used as materials to conduct high-throughput sequencing. The differentially expressed genes in midgut of the 5th instar larvae of 932 and Haoyue under high temperature( 35 ℃ ± 0. 5 ℃) and high humidity( 95% ±2%) treatment were identified and related metabolic pathways were preliminarily analyzed. The results showed that there were 552 differentially expressed genes in midgut ofthe 5th instar larvae of these two silkworm varieties under normal rearing,and these genes were divided into 44 GO functional categories. Compared with silkworm variety Haoyue which is relatively sensitive to high temperature and high humidity,the genes associated with neurite,antioxidant activity,electron carrier activity,and receptor activity were expressed at higher levels in silkworm variety 932 which is tolerant to high temperature and high humidity. Compared to the control group under normal condition,303 and 276 differentially expressed genes have been identified in midgut of the5 th instar larvae of 932 and Haoyue under high temperature and high humidity stress respectively. KEGG Pathway analysis showed that differentially expressed genes in 932 were enriched significantly in the pathways of oxidative phosphorylation,protein digestion and absorption,amino acid metabolism and glutathione metabolism etc.,and the differentially expressed genes in Haoyue were enriched significantly in the pathways including pancreatic secretion,protein digestion and absorption,bacterial and viral infection,m TOR signaling pathway and so on.
