尿中锰的直接稀释-石墨炉原子吸收光谱测定法及其不确定度分析

摘  要:目的建立一种石墨炉原子吸收光谱法测定尿锰的方法,并分析其不确定度的来源。方法尿样用超纯水直接稀释后采用石墨炉原子吸收光谱法进行测定,并且依据JJF1059—1999评定实验结果的不确定的来源。结果尿锰浓度在0~4.0μg/L范围内具有良好的线性关系,相关系数r=0.9990;方法检出限为0.08μg/L,样品加标回收率为98.1%~98.5%;相对标准偏差(RSD)为1.6%~2.9%。尿锰浓度为15.51wg/L的样品,其扩展不确定度为1.30txg/L(k=2)。结论该方法操作简便、快速、准确,避免了杂质引入干扰测定,满足生物材料中有害物质测定规范要求,可用于实验室尿锰的测定。石墨炉原子吸收光谱法测定尿锰的不确定度主要来源于绘制标准曲线和样品定量,标准溶液的配制和样品的稀释等其他几种来源对合成不确定度均有一定的贡献。[Objective ] To develop a method for determining the concentration of manganese in urine by the graphite furnace atomic absorption spectrometry (GFAAS), assess the uncertainties during the process.[Methods] The samples were diluted by the ultrapure water, and the concentration of manganese was determined by GFAAS. The sources of the uncertainties were assessed according to the JJF1059-1999. [Results] When the concentrations were between 0 and 4 μg/L, there was a good linear relationship with the correlation coefficient of 0.999 0. The detect limit was 0.08 p,g/L, the recovery of standard addition was 98.1%-98.5%, and the relative standard deviations were between 1.6% and 2.9%. The expanded uncertainty was 1.30 μg/L (k=2) when the concentration of manganese was in the urine was 15.51 μg/L. [ Conclusion ] The method is simple, rapid and accurate, which can simultaneously avoid contamination during detection and meet the requirements for the determination of hazardous substances in the biological material, and it can be used in the determination of manganese concentration in urine. The uncertainties mainly come from the processes of drawing the standard curve and quantifying samples, while preparation of standard solution and sample dilution also make contributions to the synthetic uncertainty.
