摘 要:目的:研究红芸豆植物凝集素(PHA)在终止小鼠妊娠过程中对胎盘的影响,为研究红芸豆PHA终止妊娠机制提供理论与实验依据。方法:选择妊娠14d的孕鼠,经阴道子宫注射红芸豆PHA(以注射PBS为对照),并于注射后2、5、8h及发生流产时分别收集胎盘与部分子宫进行台盼蓝染色并镜检,提取DNA并电泳,部分匀浆用ELISA试剂盒测定8异前列腺素F2α(8-iso-PGF2α)。并用SPSS 11.5软件进行独立样本t检验;同时对部分组织材料采用TUNEL试剂盒检测胎盘细胞的凋亡情况及常规染色镜检,比较PHA组与PBS组的差别。结果:1与PBS组相比较,红芸豆PHA可导致妊娠小鼠胎盘出现充血和组织坏死的现象,胎盘边缘有明显台盼蓝染色。2甲基绿-派诺宁染色与DNA电泳条带显示胎盘出现组织坏死和细胞凋亡现象。3胎盘匀浆8-iso-PGF2α水平无明显变化,而子宫匀浆8-iso-PGF2α水平明显下降。结论:注射芸豆PHA后引起小鼠胎盘组织坏死、凋亡,提示芸豆PHA终止妊娠可能是通过引起胎盘组织组织坏死、细胞凋亡或副凋亡途径达到的。Objective.. To evaluate morphological changes of cellular and tissues of uterine endometrial and placenta after administration of phytohemagglutinin (PHA) from red kidney bean for termination of middle-late pregnancy in mice. Methods: Mice with 14 days gestation age (middle-late period of pregnancy) were injected PHA from red kidney bean to one side of uterine by nonsurgical method through vagina. PBS were used in control group. Placenta and uterine were collected at 2, 5, 8 hours after administration PHA or PBS (control group). Trypan blue and methyl green-pyronin were prepared for staining and observing. The endometrial and placenta tissue were collected and prepared for micro- scopic and TUNEL analysis for apoptosis, DNA extraction and electrophoresis, homogenating and preparing for PGF2~ analysis. SPSS 11.5 software was used for data analysis. Results: Compared with the control group, PHA treatment induced blood congestion and necrosis in placenta and apoptosis as well as dye by trypan blue at the edge of the placenta. The result of methyl green-pyronin and DNA electrophoresis showed necrosis and apoptosis of placenta. The change of the level of 8-iso-PGF2α was no significant in placenta homogenate, while obvious in uterus homogenate. Conclusion: Administration of PHA could induce the placenta to apoptosis and necrosis, which may be the mechanismof PHA in termination of pregnancy.
