摘 要:目的对MTT法测定有色脂溶性化合物对癌细胞抑制作用的关键影响因素进行分析改进,并观察7种新型化合物对SMMC-7721人肝癌细胞的抑制作用。方法以SMMC-7721人肝癌细胞为目标,对MTT法测定有色脂溶性化合物的关键影响因素进行实验分析并改进;用改进后的方法测定7种新型化合物对该细胞系的抑制作用。结果药液的溶解、溶剂毒性、化合物的颜色及关键步骤的操作,对有色脂溶性化合物的MTT实验结果影响较大,应当合理进行实验设计,减弱或避免上述因素的不利影响;7种新型化合物对SMMC-7721人肝癌细胞具有不同程度的抑制作用,且随着药物浓度降低,抑制率呈下降趋势。结论本研究结果为有色脂溶性化合物的MTT实验提供了方法参考。Objective To analyze the key factors in MTT assay of lipophilic and colored compounds to improve the experimental method, and determine the inhibitory effect of 7 new compounds to SMMC-7721 human liver can- cer cells. Methods SMMC-7721 human liver cancer cells were targeted to analyze the key factors and improve the MTT assay of lipophilic and colored compounds, and then test the inhibitory effect of 7 new compounds to the cells. Results The dissolution, toxicity and color and the key steps of experiment played an important role in the MTT as- say of these compounds. We should design the experiment reasonably to weaken or avoid the adverse effects. Seven compounds showed different inhibitory effects on SMMC-7721 human liver cancer cells, and the inhibition decreased with dose. Conclusion The study provides a reference for the MTT assay of lipophilic and colored compounds.
