摘  要:目的探讨体外培养的心脏干细胞巢的微观结构。方法取8周龄SD大鼠心室肌切成1mm3组织块,经胰酶和胶原酶消化后体外培养,待有心脏干细胞长出后取组织块行透射电镜检查,观察其超微结构。取第3代心脏干细胞行美兰染色及JanusgreenB染色观察。结果心脏干细胞巢散在分布于崩解的心肌细胞问,由Telocytes细胞、细胞外基质和干细胞共同构成。体外培养的心脏干细胞间散在分布数量不等的Telocytes细胞。结论Telocyte细胞参与体外培养的心脏干细胞巢的构建,体外培养的心肌组织块可作为观察研究心脏干细胞增殖、迁移、分化及其生长微环境的良好模型。Objective To explore the microstructure of cardiac stem cell niches cultured in vitro. Methods The ventricular myocardium were obtained from Sprague Dawley rats aged 8 weeks and were cut into fragments less than 1 mm3. Following partial digestion with trypsin and collagenase, specimens were incubated in vitro. When a layer of stromal - like cells emerged from the cardiac explant, the explant was harvested for electron microscopic studies. The third generation of cardiac stem cells were stained by Meth- ylene blue or Janus green B for examination. Results Cardiac stem cell niches scattered in broken - down myocardium and were composed by stem cells, Telocytes and extracellular matrix. Different amount of Te- locytes were also observed scattered between the third generation of stem cells. Conclusion Telocytes are involved in construction of adult cardiac stem cell niches cultured in vitro. The cardiac explant may be used as a good pattern for observation and study of cardiac stem ceils' proliferation, migration, differentiation and the microenvirenment for their growth.
