摘 要:通过使用去甲基化试剂5-氮-2'脱氧胞苷(5-aza-2'deoxycytidine,5-Aza-CdR)和低氧处理小鼠,从而揭示低氧和5-Aza-CdR对蛋白磷酸酶1γ(protein serine/threonine phosphatase-1γ,PP1γ)表达的影响以及对小鼠学习记忆的影响。水迷宫筛选小鼠,正常小鼠随机分为对照组和试验组2组,分别给小鼠大脑立体定位注射10g/L BSA和10μmol/L 5-Aza-CdR至右侧脑室,休息1周,水迷宫测试小鼠学习记忆能力情况,然后依次处死低氧后3、2、1、0d的小鼠。最后通过Real-time PCR检测蛋白磷酸酶1的转录水平的表达。5-Aza-CdR和低氧分别处理小鼠都会提高小鼠的学习记忆能力,抑制PP1γmRNA的转录水平,但是联合处理对小鼠的PP1γmRNA转录水平却没有影响。说明5-Aza-CdR和低氧抑制PP1γ的转录水平,促进小鼠海马区突触可塑性的形成。The aim of this study was to detect the effect of 5-aza-2'deoxycytidine (5-Aza-CdR) and hypoxia tolerance on protein phosphatasely,and then explore the relationship between the memory and learning in mice. The mice were screened by water maze,the ordinary mice were randomly divided into groups:control mice injected with 10 g/L BSA and experimental mice injected with 10 μmol/L 5 μL 5-Aza-CdR,and then they were tested in water maze,and treated by hypoxia,and last they were euthanasia after one/two/there day hypoxia. The hippocampis were taken,and the mRNA levels of protein phosphatase-17 were measured. The results showed that 5-Aza-CdR and hypoxia can up-regulated the ability of learning and memory, down-regulated the PP17 mRNA level, but unchanged if 5-Aza-CdR and hypoxia acted on mice if them were treated at the same time. PP17 mRNA levels were inhibited by 5-Aza-CdR and hypoxia, the formation of synaptic plasticity was promoted.
