摘 要:目的寻找可预测多发性肌炎(PM)及合并肺间质病变(ILD)发生、发展的血清蛋白质。方法收集PM、PM+ILD患者及正常人(对照组)各3例,用双向电泳分离血清蛋白质点。凝胶经考马斯亮兰染色后,采用Analysis 2D7.0软件进行分析,光密度差异(ratio)>2且差别具有统计学意义(P<0.05)的蛋白质点为差异表达蛋白。基质辅助激光解析电离飞行时间质谱进行鉴定;ELISA方法进行验证。结果通过2-DE图谱分析发现,15个差异表达蛋白质点,质谱鉴定出9种蛋白质。与对照组比较,PM组表达上调的蛋白为血清淀粉样蛋白A、载脂蛋白A1、凝集素、血清白蛋白;PM组表达下调的蛋白为转铁蛋白(TRF);PM+ILD组表达下调的蛋白为锌-a2-糖蛋白、TRF、a2-H2-糖蛋白、a1-β-糖蛋白。与PM组比较,PM+ILD组表达下调的蛋白为TRF、β2-糖蛋白1。ELISA进一步验证显示,TRF在各组间表达差别无统计学意义(P>0.05)。结论 PM组、PM+ILD组及对照组3组间存在8种差异表达的蛋白,这些蛋白可能是PM及ILD发病的相关蛋白,参与了疾病的进程。Objective To search the serum proteins with differentially expression in patients with polymyositis (PM ) with or no with Interstitial Lung Disease (ILD) . Methods 3 PM patients ,3 PM+ILD patients and 3 healthy controls were included in our study . The proteins in serum were separated by two‐dimensional electrophoresis . The proteins with significant differential expression (ratio〉2 and ano‐va≤0 .05) among three groups were identified by matrix‐assisted laser desorption/ionization tandem time‐of‐flight mass spectrometry (MALDI‐TOF‐MS) .The results were certified by ELISA . Results We identified 9 proteins from 15 differential protein points by MS . Compared with healthy controls ,4 dif‐ferential proteins espressed significantly highly that amyloid A protein , apolipoprotein A1 ,Serum albu‐min ,and Clusterin ,but Serotransferin (TRF)significantly declined in the PM patients . Compared with healthy controls , the expression of 4 differential proteins that serotransferrin , zinc‐a2‐glycoprotein , a2‐H2‐glycoprotein ,a1‐beta glycoprotein significantly declined in the PM+ILD patients . Compared with PM group ,the expression of β2‐glycoprotein 1 and serotransferrin significantly decreased in the PM+ILD patients . We found that the expression of TRF has no difference in all groud by ELISA . Conclusion There were 8 kinds differential espressed protein among PM ,PM + ILD and healthy controls ,and these proteins were likely to be the pathogenetic protein of polymyositis involve in progress of the disease .
