摘 要:目的观察虎掌草醇提物的抗肿瘤活性及对荷瘤小鼠免疫功能的影响,并对其安全性进行初步评价。方法改良MTT法检测虎掌草醇提物对人Hep G-2、K562、A549和PC-3细胞株的增殖抑制活性;采用小鼠移植性肿瘤模型,研究虎掌草醇提物对小鼠肝癌H22生长的影响;免疫器官称重法检测对荷瘤小鼠免疫器官指数的影响;ELISA法检测荷瘤小鼠血清IL-2、TNF-a水平;小鼠急性毒性试验初步评价虎掌草醇提物的安全性。结果虎掌草醇提物在体外对人肿瘤细胞株具有明显的增殖抑制活性,其IC50值均小于10μg/m L;虎掌草醇提物1.0、2.0 g/kg剂量可明显抑制小鼠肝癌H22的生长,同时对小鼠体重无明显影响;虎掌草醇提物可明显提升小鼠胸腺指数和脾脏指数,可明显提升小鼠血清TNF-α和IL-2水平。虎掌草醇提物小鼠灌胃给药LD50为6.218 g/kg,95%可信限为5.104 g/kg^7.526 g/kg,其毒性可能涉及的组织、器官、系统为中枢神经、神经肌肉、自主神经和肺脏功能损伤。结论虎掌草醇提物具有明显的体内、外抗肿瘤活性,其活性可能与增强动物免疫力,提升小鼠血清IL-2、TNF-α水平有关。Objective: To observe the anti - tumor activity of alcohol extraction of Brook Anemone and its effect on the immunity of tumor - bearing mice. Methods : MTr was modified to detect alcohol extract of Brook Anemone on the proliferation inhibitory activity of Hep G - 2, K562-,- AM9 and PC - 3 cell line of people. Tumor transplanted mice model was used to study the influence of the alcohol extract of Brook Anemone on the growth of liver cancer H22. Immune organ weight was used to detect immune organ index of the tumor - bearing mice ; ELISA method to detect their serum IL - 2 and TNF - a levels ; the acute toxicity test preliminarily to evaluate the safety of Brook A- nemone. Results : The alcohol extract of Brook Anemone had a significant growth inhibitory activity in vitro for human tumor cell lines and its ICso values were less than 10 μg/mL. The 1.0, 2. 0 g/kg dose of alcohol extract significantly inhibited H22 liver cancer growth of the mice, but no obvious effect on body weight of the mice. Brook Anemone can significantly raise the thymus index and spleen index and the serum TNF - ct and IL - 2 levels. When the mica were given intragastric infusion of alcohol extract LDS0 6. 218 g/kg, 95% confidence in- terval was 5. 104 g/kg ~ 7. 526 g/kg. Its tissue toxicity may be involved in the central nervous, neuromuscular and autonomic nerve injury and lung function impairment of organs, central nervous system and nerves muscle. Conclusion : The ethanol extract of Brook Anemone has obvious anti - tumor activity in vivo and in vitro and its activity may be related to the improvement of animal immunity and the serum IL - 2 and TNF - α level of the mice.
