摘 要:目的:观察电针内关穴对心肌缺血大鼠缺氧诱导因子(HIF-1α)及酸敏感离子通道(ASIC2、ASIC3)基因表达的影响。方法:SD大鼠45只随机对照方法分为正常组、模型组、内关组、列缺组、非经非穴组,每组10只。异丙肾上腺素造模成功后,分别进行电针治疗。1周后,Realtime RT-PCR法检测各组左心室心肌ASIC2、ASIC3的基因表达,ELISA法检测各组血清HIF-1α含量。结果:电针内关穴可降低心肌缺血大鼠ASIC2、ASIC3基因表达,降低血清中HIF-1α含量,与模型组比较有显著性差异(P<0.01),且优于列缺组与非经非穴组(P<0.01)。结论:电针内关穴可降低血清中HIF-1α含量,下调心肌缺血大鼠ASIC2、ASIC3基因表达,从而抑制酸敏感离子通道开放,有效改善心肌缺血大鼠心肌细胞的损伤。Objective: To observe the influence of electroacupuncture (EA) at Neiguan (PC 6) on expression of hypoxia inducible factor-1 alpha (HIF-1 α ) and acid sensing ion channel-2,3 (ASIC2, ASIC3) in rats with myocardial ischemia. Methods: 45 SD rats were randomly divided into control group (group A), model group (group B), Neiguan (PC 6) group (group C), Lieque (LU 7) group (group D) and sham acupoints group (group E), with 10 rats in each group. The rat models were established by multipoint subcutaneous injection with isoprenaline (85mg/kg) in 4 limbs once per day. After establishing of rat models, the rats in each group were treated with EA for a week. All the rats were narcotized by intraperitoneal injecting with chloral hydrate (lmL/100g), and the left ventricular myocardium of rats was separated and kept in cryogenic refrigerator. Genetic expression of ASIC2 and ASIC3 was determined by using RT-PCR method. The content of HIF-1 α in serum was tested by using ELISA method. Results: EA at Neiguan (PC 6) could decrease the genetic expression of ASIC2 and ASIC3 and the content of HIF-1 α in serum, and the difference was significant compared with model group (P〈0.01). Conclusion: EA at Neiguan (PC 6) could inhibit the opening of acid sensing ion channels and improve the myocardial cell injury through decreasing the content of HIF-1 α in serum and down-regulating the genetic expression of ASIC2 and ASIC3.
