十精丸含药血清对光老化ESF-1细胞分泌MMP-1、TIMP-1的影响

摘　　要：目的:研究十精丸含药血清对光老化人皮肤成纤维细胞(ESF-1)分泌MMP-1、TIMP-1的影响。方法:制备十精丸含药血清,采用UVA照射ESF-1细胞建立光老化模型,照射后用含不同浓度十精丸的大鼠血清进行干预,24h后采用ELISA方法检测各组细胞分泌MMP-1、TIMP-1的含量。结果:与正常对照组比较,模型对照组细胞活力降低、MMP-1表达升高、TIMP-1表达降低(P<0.01);与模型对照组比较,十精丸中、高剂量含药血清组细胞活力升高、MMP-1表达降低、TIMP-1表达升高(P<0.05,P<0.01)。结论:十精丸含药血清通过调节MMP-1、TIMP-1的表达来拮抗UVA对皮肤成纤维细胞的损伤。Objective： To investigate the influence of serum containing Shijing Pill on expression of MMP-1 and TIMP- 1 secreted by photoaging ESF-1 cells. Methods： The photoaging ESF-1 cell model was established by using UVA irradiation, and the cells were intervened with rat serum containing Shijing Pill after cell model establishing. After intervening for 24h, the content of MMP-1 and TIMP-1 was determined by using Eslisa method. Results： Compared with control group, the expression of MMP-1 in cells of model group was increased and the expression of TIMP-1 in cells of this group was decreased （P〈0.01）. Compared with model group, the expression of MMP-1 in cells of middle and high dosage of serum containing Shijing Pill groups was decreased, and the expression of TIMP-1 in cells of these groups was increased （P〈0.05, P〈0.01）. Conclusions： Serum containing Shijing Pill could resist the damages in skin fibroblasts caused by UVA through regulating on expression of MMP-1 and TIMP-1 .
