摘  要:目的:探讨地五养肝胶囊(DWYG)对肝纤维化大鼠肝脏Hedgehog(Hh)信号通路的影响。方法:Wistar大鼠随机分为空白对照组、肝纤维化模型组和DWYG干预组,四氯化碳(CCl4)诱导肝纤维化模型。应用HE和Masson染色检测不同处理组大鼠肝组织损伤和纤维化程度;应用Western Blot和Real-time q RT-PCR分别检测不同处理组大鼠肝组织Hh信号通路的蛋白和基因表达水平。结果:与空白对照组相比,肝纤维化模型组大鼠出现明显肝组织损伤和纤维化改变,且纤维化肝组织中Shh、Gli1、Smo和Ptc的蛋白和基因表达水平显著上调(P<0.05);而与肝纤维化模型组相比,DWYG干预可显著减轻肝组织损伤和纤维化程度,同时显著下调纤维化肝组织中Shh、Gli1、Smo和Ptc的蛋白和基因表达水平(P<0.05)。结论:DWYG抗纤维化效应与其抑制纤维化大鼠肝组织中过度活化的Hh信号通路相关。Objective: To investigate the impacts of Diwu Yanggan Capsule (DWYG) on Hedgehog (Hh) signal pathway in the liver tissues of CC14-induced fibrotic rats. Methods: Wistar rats were randomly divided into blank control group, liver fibrosis model group and DWYG-treated group. Liver fibrosis was established in rats by CC14 induction. HE and Masson stain were carded out to examine the effects of DWYG on liver injury and fibrosis degree. The impacts of DWYG on the mRNA and protein expression levels of Hh signal pathway were further examined by Real-time qRT-PCR and Western Blot analysis, Results: Compared with blank control group, rats in liver fibrosis model group had remarkable liver injury and fibrosis, and the mRNA and protein expression levels of Shh, Glil, Smo and Ptc were obviously up-regulated (P〈0.05). Moreover, compared with liver fibrosis model group, the degrees of liver injury and fibrosis in DWYG-treated group were significantly alleviated, and the mRNA and protein expression levels of Shh, Glil, Smo and Ptc were markedly down-regulated (P〈0.05). Conclusion: The anti-fibrotic effect of DWYG is associated with inhibiting the excessive activation of Hh signaling pathway in CC14-induced fibrotic liver tissues.
