参附注射液对“失血性休克-内毒素”二次打击急性肺损伤治疗作用研究

摘　　要：目的:探讨参附注射液对"失血性休克-内毒素"二次打击急性肺损伤大鼠的治疗作用及机制。方法:以雄性Wistar大鼠为对象通过颈动脉放血+气管脂多糖(LSP)注射建立大鼠急性肺损伤模型,并通过静脉注射参附注射液予以治疗,以正常Wistar大鼠作为对照。术后通过病理检测、电镜观察肺超微结构变化。ELISA法检测肺脏中肿瘤坏死因子(TNF-α)和白细胞介素-6(IL-6)表达变化。结果:经病理和ELISA检测显示参附注射液组肺脏损伤情况显著轻于急性肺损伤组(ALI组)(P<0.05);ELISA法检测发现急性肺损伤组大鼠肺脏TNF-α和IL-6表达较空白对照组显著升高(P<0.05),参附注射液组较急性肺损伤组显著降低(P<0.05)。结论:参附注射液可以减轻大鼠急性肺损伤,其机制可能是通过抑制肺脏中TNF-α和IL-6的表达。Objective： To investigate the effects and mechanism of Shenfu （SF） Injection on two-bit with hemorrhagic shock and endotoxin on acute ALI rats. Methods： Male Wistar rats blooded through carotid artery and injected LSP into trachea were used to establish ALl models. These rats were treated by injecting SF Injection into venous. Normal Wistar rats were used as control. Curative effect of SF Injection was evaluated through pathological examination and electron microscope. TNF- α and IL-6 expression in lung was tested by ELISA. Results： After SF Injection treatment, pathological examination and ELISA test results showed lighter lung injury compared with acute lung injury rats （P〈0,05）. ELISA results showed TNF- α and IL-6 expression increased in rats with acute lung injury and could decrease significantly after SF Injection treatment （P〈0.05）. Conclusion： SF Injection could alleviate acute lung injury. The mechanism might be due to inhibiting TNF-α and IL-6 expression in lung.
