鳖甲煎丸对肝癌细胞周期及Wnt/β-catenin信号通路中β-catenin、COX-2蛋白表达水平的影响

摘　　要：目的:研究鳖甲煎丸对肝癌细胞Hep G2细胞周期、Wnt信号通路中β-catenin蛋白及其磷酸化水平、靶基因COX-2蛋白表达水平的影响,探讨鳖甲煎丸抗Hep G2细胞增殖、黏附与侵袭的作用机制。方法:运用血清药理学方法,以鳖甲煎丸含药血清、空白血清及索拉菲尼含药血清分别培养Hep G2细胞,采用流式细胞术、Western Blot检测药物对细胞周期、Wnt信号通路中β-catenin、phospho-β-catenin(Ser675)、COX-2蛋白表达水平的影响。结果:鳖甲煎丸高中剂量组、索拉菲尼可使Hep G2细胞发生G1期阻滞,下调Hep G2细胞中β-catenin、COX-2、phospho-β-catenin(Ser675)蛋白表达水平。结论:鳖甲煎丸可抑制Hep G2细胞增殖、黏附与侵袭,其机制可能与其使细胞发生G1期阻滞、减少β-catenin在细胞中的积聚、干预phospho-β-catenin(Ser675)的促转录作用、抑制靶基因COX-2表达具有相关。Objective： To research the influence of Biejiajian Pill on cell cycle and expression of β-catenin and COX-2 of Wnt/β-catenin signaling pathway in hepatoma carcinoma （HepG2） cells in order to investigate the mechanism of Biejiajian Pill （BP） in inhibiting the proliferation, adhesion and invasion of HepG2 cells. Methods： HepG2 cells were cultured in serum containing BP, blank serum and serum containing Sorafenib separately by using serologic pharmacological method. The cell cycle of HepG2 was measured by using flow cytometry. The expressions of β-catenin, phospho-β-catenin （Ser675） and COX-2 were measured by using Western Blot method. Results： High and middle dosage of serum containing BP and Sorafenib could block the cell cycle at G1 phase and decrease the expression of β-catenin, phospho- β -catenin （Ser675） and COX-2. Conclusion： BP could effectively inhibit the growth and proliferation, adhesion and invasion of HepG2 cells by blocking the cell cycle, decreasing the accumulation of β-catenin in cells, intervening the effect of Ser675 on promoting transcription and inhibiting expression of COX-2 in the Wnt/ β-catenin signaling pathway.
