骨蚀灵胶囊对兔早期激素性股骨头坏死骨组织VEGF mRNA表达的影响

摘　　要：目的:观察骨蚀灵胶囊对兔早期激素性股骨头坏死(SANFH)骨组织中VEGFmRNA表达的影响,探讨中药防治早期激素性股骨头坏死的可行性及其机理。方法:选用成年日本大耳白兔,随机分为3组:正常对照组、模型组和中药组。采用马血清和甲强龙联合应用的方法复制SANFH模型,中药组于激素注射完毕后第二天进行骨蚀灵胶囊灌胃治疗,4、8周分批处死动物,应用RT-PCR技术检测股骨头骨组织中VEGFmRNA表达的变化。结果:与正常组比较,模型组股骨头骨组织中VEGFmRNA表达明显降低,8周时较4周时降低明显,差异有统计学意义(P<0.05);中药组VEGFmRNA表达较模型组明显升高,但仍未达到正常水平,差异有统计学意义(P<0.05)。结论:增强股骨头骨组织VEGFmRNA表达是骨蚀灵胶囊有效治疗激素性股骨头坏死的作用机制之一。Objective： To investigate the prevention and treatment effect of Chinese medicine Gushiling capsule on the early- stage steroid- induced necrosis of femoral head,through determining the expression of VEGF mRNA in femoral head. Methods： Healthy adult Japanese big ear rabbits were selected,which were randomly divided into normal control group,model group and Chinese medicine（ Gushiling） group. SANFH models were made by injected horse serum and methyprednisolone,then from the next day,the rabbits of the Chinese medicine group were given Gushiling capsule lavage,the other two groups were given equal volume of distilled water lavage. At the 4th,8th week,the rabbits of the three groups were slaughtered and the VEGFmRNA were quantitatively analyzed by RT- PCR. Results： To compare with normal group,the VEGFmRNA expression of model group was significantly lowered,that was significantly lower in the 8th week than that in the 4th week and the differences was obviously（ P〈0. 05）; the VEGF mRNA expression of Chinese medicine group was significantly higher than that in model group but did not reach the normal level and the differences was obviously（ P〈0. 05）.Conclusion： Raising the VEGF mRNA expression on bone of femoral head is one of the mechanism that Gushiling capsule effectively treat early steroid- induced necrosis of femoral head
