摘 要:目的:比较非小细胞肺癌脾虚痰湿证和其他证型血清蛋白质组学的差异,筛选能用于非小细胞肺癌脾虚痰湿证客观化辨证的血清生物学标志物。方法:利用双向荧光差异凝胶电泳技术(two-dimensional fluorescence difference gel electrophoresis,2D-DIGE)比较非小细胞肺癌脾虚痰湿证血清(3例)和其他证型血清(3例)蛋白图谱的差异,使用基质辅助激光解吸电离—飞行时间质谱(matrix-assisted laser desorptionionization time-of-flight mass spectrometry,MALDI-TOF-MS)及生物信息学相关技术对获得的差异蛋白进行鉴定和初步分析。结果:成功建立了非小细胞肺癌脾虚痰湿证血清差异蛋白质组学的研究方法;通过质谱分析初步鉴定出6个差异蛋白。结论:非小细胞肺癌脾虚痰湿证血清和其他证型血清蛋白表达存在明显差异,可能为非小细胞肺癌脾虚痰湿证辨证客观化提供新的线索和实验基础。Objective: To compare the proteomics study on the non-small cell lung cancer with syndrome of spleen deficiency and phlegm- damp with other syndromes, to select the serum biolandmarks for non-small cell lung cancer with syndrome of spleen deficiency and phlegm-damp. Methods: Two-dimensional fluorescence difference gel electrophoresis (2D-DIGE) was applied to analyze the sera ob- tained from the patients with spleen deficiency and phlegm-damp (3 cases) and patients with other syndromes (n = 3 cases). The differ- entially expressed proteins were identified by matrix-assisted laser desorption ionization time-of-flight mass spectrometry( MALDI-TOF- MS). Results : Comparative proteomic data of serum from the patients with splenic asthenia and phlegm-damp was successfully obtained. Six differentially expressed protein spots were observed. Conclusion:There are significant differences between serum proteins obtained from the patients with syndrome of splenic asthenia and phlegm-damp and patients with other syndromes, might be utilized as potential biomarkers for the diagnosis and treatment for the syndrome of splenic asthenia and phlegm-damp.
