肾消康对糖尿病骨质疏松大鼠骨胶原重塑的影响

摘　　要：目的:观察肾消康对糖尿病性骨质疏松模型大鼠骨胶原重塑的影响。方法:雄性Wistar大鼠73只,随机选15只为空白组,以普通饲料喂养,经左下腹注射柠檬酸缓冲液25 mg·kg-1。其余58只称取质量,以25 mg·kg-1链脲佐菌素左下腹腔注射,造模成功后再随机分为模型组、治疗组,两组均喂以高热量饲料13周,治疗组以中药复方肾消康灌胃;空白组、模型组以生理盐水灌胃,均1次·d-1。第26周末处死大鼠,取胫骨、腰椎骨制备扫描电镜标本,并对其进行形态学观察。结果:模型组胫骨骨胶原大面积流失,排列紧密度降低、走行散乱、排列不规整,甚至个别视野下骨胶原组织出现断裂、塌陷现象;腰椎骨骨胶原完全消失、骨基质完全暴露。肾消康治疗组大鼠骨胶原组织恢复明显,尤其是在腰椎骨体现更为显著,骨的力学结构得到改善,骨胶原丢失降低,排列更紧密、有规律,促进了骨胶原组织的再生与重塑。结论:肾消康可有效促进骨胶原组织的再生与重塑,从而为有效防治糖尿病骨质疏松症提供了可行的思路与方法。Objective ：To observe the effects on the remodeling of rats ＇ossein of diabetes with osteoporosis by Shenxiao Kang. Methods： 73 Wistar rats with male were randomly selected blank group （ 15 rats）, which was given common fodder and injected citrate buffer solu- tion 25 mg ·kg- 1 by left lower quadrant. Others were weighed and injected streptozotocin 25 mg · kg- 1 by left lower quadrant, after the remodeling, they were randomly divided into model group, treatment group. The two groups were given high heat fodder for 13 weeks, the treatment group was gavage by Chinese herbal compound Shenxiao Kang, the blank group and the model group was gavage by normal sa- line, 1 time a day. After the 26weeks, executed the rats, the tibia, preparation（ scanning electron microscopy sem） specimens, and having morphological observation. Results：In the model group, the tibia bone collagen had lossed with large area, close degree was at lower line and messy, not neat, even the perspective of individual collagen tissue appear rupture and collapse phenomenon, collagen disappear com- pletely, bone matrix was exposed completely. Renal elimination. In the treatment group,rats collagen tissue recovery significantly, espe- cially in lumbar vertebra,improving the mechanical structure, reduce the ossein, arrangement was more close and regularly, promote the regeneration and remodeling. Conclusions ：Shenxiao Kang can promote effectively the regeneration and remodeling of ossein ,which pro- vides the effective prevention and treatment of diabetic osteoporosis feasible idea and method.
