醒鼻凝胶剂对变应性鼻炎豚鼠胸腺基质淋巴细胞生成素表达的影响

摘　　要：目的:观察醒鼻凝胶剂对变应性鼻炎(allergic rhinitis,AR)豚鼠鼻黏膜胸腺基质淋巴细胞生成素(thymic stromal lymphopoietin,TSLP)及Th2型细胞因子白介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumour necrosis factor,TNF-α)表达的影响,探讨其治疗AR的可能机制。方法:健康豚鼠60只随机分为正常对照组,模型对照组,布地奈德组,醒鼻凝胶剂高剂量组、中剂量组、低剂量组,每组10只。卵清蛋白加氢氧化铝腹腔注射制备AR豚鼠模型,醒鼻凝胶剂组给予不同剂量醒鼻凝胶剂治疗,连续11 d。ELISA法检测鼻腔灌洗液IL-6、TNF-α的表达,RT-PCR、免疫组织化学检测鼻黏膜TSLP mRNA和TSLP蛋白的表达。结果:模型组IL-6、TNF-α含量和TSLP mRNA、TSLP蛋白的表达量明显高于对照组(P<0.01);醒鼻凝胶剂组和布地奈德组IL-6、TSLP mRNA和TSLP蛋白的表达量明显低于模型组(P<0.01);醒鼻凝胶剂组和布地奈德组TNF-α明显低于模型组(P<0.05)。结论:醒鼻凝胶剂可抑制AR豚鼠的炎症反应,其机制可能通过抑制TSLP的表达,减少TSLP诱导的Th2型细胞因子IL-6和TNF-α的产生而实现。Objective：To observe the effects of Xingbi gel on the expression of thymic stromal lymphopoietin（TSLP） and Th2 type cyto- kines IL-6,TNF-α in allergic rhinitis （AR） guinea pigs. To explore the therapeutic mechanism of the Xingbi gel on AR. Methods：60 healthy guinea pigs were randomly divided into 6 groups, normal control group, model group, budesonide group, Xingbi gel group in low, middle,and high dose（n = 10 each）. The Guinea pigs were modeled to AR by intraperitoneal ovalbumin injection combined with alu- minium hydroxide,while the Guinea pigs in the Xinghi gel group were administered with respectively dose Xingbi gel for 11 consecutive days. The levels of interleukin（IL） -6 and TNF-α in nasal lavage fluid were measured by ELISA. The mRNA and protein expressions of TSLP in the nasal mucosa were measured by real-time PCR and immunohistochemistry respectively. Results ： The levels of IL-6, TNF-α the expressions of TSLP mRNA and TSLP were markedly elevated in the model group versus the normal control group（ all P 〈 0.01 ）. The levels of IL-6, the expressions of TSLP mRNA and TSLP in the Xingbi gel and budesonide groups were obviously down-regulated versus the model group（ all P 〈 0.01 ）. The levels of TNF-α in the Xingbi gel and budesonide groups were markedly decreased versus the model group （P 〈 0.05 ）. Conclusion：The Xingbi gel can alleviate the nasal inflammation in allergic rhinitis（AR） guinea pigs. The above effects may be partially due to the inhibited expressions of TSLP and the successively reduction of Th2 type cytokines IL-6, TNF- α which induced by TSLP.
