摘  要:为探讨褪黑素(MLT)对牛卵母细胞成熟、精子获能和胚胎发育的抗氧化效果。在体外培养液中添加10-9 M MLT,以评估牛卵母细胞成熟率及胚胎发育效果;同时利用目测法、低渗膨胀法、JC-1和考马斯亮兰染色法评估不同浓度MLT(0 M、10-3 M、10-4 M和10-5M)添加在牛精子获能液经上游法处理45min后对精子质量的影响。结果表明:与对照组相比,添加10-9 M MLT既可显著提高牛卵母细胞的成熟率(62.5%vs 77.8%)(P<0.05),又可明显刺激受精后胚胎体外发育的卵裂率(53.2%vs 67.6%)(P<0.05)和囊胚率(20.3%vs35.4%)。在精子获能液中添加10-4 M MLT上游处理45 min后,其精子活力(84.6%vs71.6%)、质膜完整率(88.8%vs 75.2%)、线粒体高膜电位百分率(83.0%vs 70.2%)和顶体完整率(87.0%vs 74.4%)均显著高于对照组(P<0.05)。这些结果有助于优化IVF技术体系,利于体外胚胎生产效率提高。The antioxidative effect of melatonin on sperm capacitation, maturation and embryonic development in bovine oocyte was evaluated in this study to investigate the effect of supplemented MLT in vitro culture on bovine oo- cyte maturation and embryo development. Bovine sperm supplementing with 0 M, 10-3 M, 10-4 M and 10-5 M me- latonin to the capacitation fluid and incubated by swim-up method for 45 rain were tested with, the method of visual observation, HOST, JC-1 and Coomassie. The results showed that supplementation with melatonin of 10-9 M con- centration in the culture medium resulted in a higher (P〈0. 05) maturation rate (77.8% and 62.5%), cleavage rate (67. 6% and 53.2%) and blastocyst rate (35. 4% and 20. 3%) compared with control group. Sperm samples with 10-4 M melatonin had a significant higher progressive motility (84. 6% vs 71.6%), higher percentage of plasma mem- brane integrity (88. 8% vs 75. 2%), higher percentage of spermatozoa with high rnitochondrial membrane potential (83.0% vs 70. 2%) and higher acrosome integrity (87. 0% vs 74. 4%) than those in control group (P〈0. 05). In conclusion, the results help to optimize IVF procedures and improve embryo development in vitro production.
