摘  要:细胞内三磷酸腺苷(ATP)的有效提取是利用ATP发光法检测微生物数量的关键.为判断霉菌在织物上的生长情况,以纯棉织物上接种的黑曲霉为对象,研究了霉菌胞内ATP的提取方法,对比了加热煮沸法、磷酸盐缓冲液法、三氯乙酸法、二甲基亚砜法和苯扎溴铵(BAC)法共5种ATP提取方法对黑曲霉胞内ATP的提取效率.研究结果表明,BAC法提取ATP效果最好;BAC法提取的适宜条件:BAC质量分数为0.50%,pH值为12.0,提取时间为5min.相比于传统的检测法,通过测定ATP含量判断织物上霉菌生长的方法使检测效率显著提高.Extraction of intracellular adenosine triphosphate(ATP)is the key of detecting the amount of microorganisms by ATP luminescence. In order to determine the growth of filamentous fungi on pure cotton fabric, Aspergillus niger inoculated on the cotton fabric was chosen as the experimental mode to study the ATP extraction methods. Five ATP extraction methods were compared, including boiling method, phosphate buffer method, trichloroacetic acid method, dimethyl sulfoxide method and benzalkonium chloride (BAC)method. The results showed that BAC method was the best method among five extraction methods and optimal conditions included 0.50%(mass fraction) BAC, pH value 12.0 and 5 min extraction time. Compared to the traditional detection method, the testing efficiency by determining the amount of ATP to determine the growth of filamentous fungi was significantly increased.
