摘 要:为研究地塞米松(DEx)对高致病性猪繁殖与呼吸综合征病毒(HP—PRRSV)感染仔猪造成的免疫系统和呼吸系统损伤的影响,本实验在断奶仔猪感染HP-PRRSVHuN4株前后,肌肉注射不同剂量的DEX。观察临床症状,检测其免疫系统相关指标的变化。结果显示:不同剂量DEX注射组实验猪均表现典型临床症状,虽然其外周血中的促炎性细胞因子IL-1β、IL-6及TNF—d水平降低,CD4+CD8‘T淋巴细胞亚群比例较HuN4单独感染组升高,CD8+CD4-T细胞比例较HuN4单独感染组降低,但DEX注射组仔猪体温始终维持在40℃-42℃,肺部、胸腺和淋巴组织病变加重,死亡率达到100%(10/10),而HuN4单独感染组仔猪死亡率仅为25%(1/4)。表明DEX对HP—PRRSV感染的断奶仔猪虽具有一定的抑制炎症反应作用,但加重了HP-PRRSV感染的不利影响。本研究为全面防控HP—PRRSV提供新的实验依据。To investigate effect of dexamethasone (DEX) on the pathogenicity of highly pathogenic porcine reproductive and respiratory syndrome virus (HP-PRRSV) in piglets, different doses of DEX were injected in piglets before (2 mg/kg-weights) and after (0.5 and 2 mg/kg-weights) artificially infecting with HP-PRRSV HuN4 strain. The results showed that all the infected piglets injected with DEX developed the typical clinical signs, and the rectal temperatures of these piglets were continuously higher within 40 ℃ to 42 ℃, which were more severe than that of pigs in only infected with virus control group. However, the levels of proinflammatory cytokines IL-1β, IL-6, TNF-α and lymphocyte subsets in peripheral blood changes were mild compared to the piglets in only infected with virus group. Eventually, the mortality was 100% (10/10) for the infected piglets injected with DEX, but the mortality was only 25% (1/4). These results demonstrated that DEX was able to improve host anti-inflammatory during HP-PRRSV infection of HP-PRRSV, but still had a strong impact resulting in lethalside effect to piglets. These data provided the basis for furtherstudy on the pathogenicity of HP-PRRSV.
