摘 要:目的:探讨不同干预手段的骨髓间充质干细胞(Bone marrow mesenchymal stem cells,BMSCs)移植术治疗肝硬化大鼠的治疗作用。方法:取肝硬化大鼠模型40只,随机分为对照组、BMSCs门静脉移植组、BMSCs门静脉移植联合注射集落刺激因子组、BMSCs门静脉移植联合济生肾气汤灌胃组,每组各10只。治疗后每周分别取血测定肝功能和肝纤维化指标,4周后处死所有大鼠。所有死亡的大鼠肝脏组织切片进行HE和Masson染色,检测大鼠肝细胞组织损伤和纤维化情况。结果:肝功能、肝纤维化、病理组织分级评分的比较上:三组移植组与对照组比较,具有统计学学意义(P<0.05);BMSCs配合注射G-CSF移植组、联合济生肾气汤移植组与单行BMSCs移植相比,具有统计学学意义(P<0.05);联合济生肾气汤移植组与配合注射G-CSF移植组相比,具有统计学意义(P<0.05)。结论:不同干预手段的BMSCs移植术对大鼠肝功能、肝纤维化程度、病理组织学等方面均有改善作用,其中联合济生肾气汤的BMSCs移植组作用最优,比单行BMSCs移植组和配合注射G-CSF移植组更能有效促进肝脏组织的修复。Objective: To observe the therapeutic effect of bone marrow mesenchymal stem cells ( BMSCs ) transplantation by different means of intervention on cirrhotic rats. Methods : 40 cirrhotic rats were randomly divided into control group, BMSCs by portal vein transplantation group, BMSCs by portal vein transplantation group combined with injection of colony stimulating factor, BMSCs by portal vein transplantation group combined with Jisheng Shenqi decoction, each group of 10 cirrhotic rats.In blood samples which collected every week were measured indicators of liver function and liver fibrosis, all the rats were killed after four weeks. The liver tissue of all of death rat slices prepared for HE and Masson staining, detection of liver tissue damage and fibrosis conditions. Results : Comparing the liver function, hepatic fibrosis and histopathological grading score : three groups of transplantation compare with the control group, have the statistically significant ( P〈O.05 ).BMSCs trans-plantation group combined with injection of G-CSF and BMSCs transplantation group combined with Jisheng Shenqi decoction were compared with single BMSCs transplantation, have the statistical significance ( P〈O.05 ); the transplantation group combined with Jisheng Shenqi decoction was compared with the group combined with injection of G-CSF, has the statistical significance ( P〈O.05 ) .Conclusion : Bone marrow mesenchymal stern cells transplantation by different means of intervention have postive influence on the change of the liver function, hepatic fibrosis and histopathological grading score. BMSCs transplantation group combined with Jisheng Shenqi decoction works best among all groups, which can more effectively promote liver tissue than the single BMSCs transplantation group or the group combined with injection of G-CSF.
