摘 要:目的观察针刺对支气管哮喘大鼠脑、肝、肾组织表面活性物质蛋白(surfactant protein,SP)-A mRNA表达的影响,探讨支气管哮喘从肺外脏腑论治的思路。方法将40只SD大鼠随机分为空白组、模型组、针刺肺组、针刺肠组及针刺肺肠组5组。采用腹腔注射卵蛋白致敏,2周后尾静脉注射卵蛋白激发支气管哮喘发作对模型组、针刺肺组、针刺肠组及针刺肺肠组进行造模;空白组在相同时间点注射0.9%氯化钠注射液。针刺肺组取尺泽、孔最、列缺、肺俞针刺;针刺肠组取曲池、合谷、天枢、上巨虚针刺;针刺肺肠组取孔最、列缺、曲池、天枢针刺。检测各组脑、肝、肾组织SP-A mRNA的表达情况。结果各组脑、肝、肾组织中均有SP-A mRNA表达。空白组、针刺肺组、针刺肠组及针刺肺肠组脑组织中SP-A mRNA的表达水平均较模型组升高,差异有统计学意义(P<0.01)。针刺肺肠组脑组织中SP-A mRNA表达水平高于针刺肠组,差异亦有统计学意义(P<0.05)。各组肝组织及肾组织SP-A mRNA表达水平比较差异无统计学意义(P>0.05)。结论支气管哮喘大鼠脑组织中SP-A mRNA发生异常表达,针刺肺经腧穴与大肠经腧穴均可对脑组织SP-A mRNA的表达产生影响。针刺对支气管哮喘大鼠肝、肾组织中SP-A mRNA表达的影响不明显。针刺治疗支气管哮喘,调神之法任何病程均可应用,从肝肾论治并不适用于新发哮喘。Objective To observe expression effect of acupuncture treatment on the surface protein ( SP)-A mRNA expression of brain, liver and kidney tissues in asthmatic rats, and to explore the approach of bronchial asthma from the exter-nal and external organs of the lungs. Methods 40 SD rats were randomly divided into blank group, model group, lung acu-puncture group, intestinal acupuncture group, lung and intestinal acupuncture group. Methods of observation of model group, lung acupuncture group, intestinal acupuncture group, lung and intestinal acupuncture group were proceeded through the in-jection of ovalbumin sensitization in the abdomen cavity and after 2 weeks, intravenous injection of the ovalbumin was carried out to induce asthma. Method of observation of blank group was proceeded through injection 0. 9% sodium chloride injection at the same time points of other groups. Lung acupuncture group selected the following acupoints: Chize ( LUS ) , Kongzui (LU6), Lieque (LU7), Feishu (BL13). In large intestine acupuncture group, the following acupoints were selected:Quchi (LIll), Hegu (LI4), Tianshu (ST25), Shangjuxu (ST37) were selected. In lung and large intestine acupuncture groups, the following acupoints were selected, Kongzui (LU6), Lieque (LU7), Quchi (LIll), Tianshu (ST25). The expression of SP-A mRNA in brain, liver and kidney tissues was observed after the rats were killed. Results The expression of mRNA SP-A was found in brain, liver and kidney in each group. The expression of mRNA SP-A in the brain tissues of the blank group, the acupuncture group, the acupuncture group and the acupuncture group were significantly higher than that of the model group (P〈0. 01). The levels of SP-A mRNA in brain tissues of rats with acupuncture group were significantly higher than that in the acupuncture group (P〈0. 05). There was no significant difference in SP-A mRNA expression in liver and kidney tissues between the three groups (P〉0. 05). Conclusion
