摘  要:为研究大花蕙兰(Cymbidium hybridum)的高频快繁再生体系,以大花蕙兰无菌原球茎、分化苗为材料,应用正交试验进行不同激素和添加物的优化筛选。结果表明:在1/2 MS+0.3 mg/L KT+0.5 mg/L NAA+25 g/L蔗糖+7 g/L琼脂+1 g/L活性炭的培养基上大花蕙兰的原球茎增殖效果最好,新增原球茎呈深绿色、形状饱满,增殖倍数达11倍;在1/2 MS+0.1 mg/L6-BA+0.4 mg/L NAA+25 g/L蔗糖+7 g/L琼脂+1g/L活性炭的培养基上茎叶分化效果最好,分化丛生芽数量最多、颜色深绿色、生长健壮,分化倍数达6.1倍;在1/2 MS+0.2 mg/L GA3+1.0mg/L NAA+0.2 mg/L IBA+25 g/L蔗糖+7 g/L琼脂+1 g/L活性炭的培养基上生根效果最好,生根数最多,平均生根数达4条,根系生长强健。In order to study the high frequency rapid propagation regeneration system of Cymbidium hybridum ,taking the fourth sterile protocorm and differentiated seedlings as materials, orthogonal test was used to optimization screen different hormones and additives.Results showed that 1/2 MS+ 0.3 mg/L KT+0.5 mg/L NAA+25 g/L sucrose +7 g/L agar+l g/L activated carbon on the culture medium of big fourth protocorm proliferation effect best, new protoeorm was dark green ,shape and proliferation ratio up to 11 times.In 1/2 MS+0.1 mg/L 6-BA +0.4 mg/L NAA +25 g/L sucrose +7 g/L agar+l g/L activated carbon medium stalk leaf differentiation effect was best, the differentiation of multiple shoot clumps had the largest number, color was dark green ,growth ,differentiation ratio reached 6.1 times.In 1/2 MS+0.2 mg/L GA3+I.0 mg/L NAA+0.2 mg/L IBA+25 g/L sucrose+7 g/L agar+l g/L activated carbon on the culture medium of rooting effect was best, root number was the most which reached 4 on average, and grew strong.
